Abstract

A capillary electrophoresis system includes a capillary reservoir. The capillary reservoir includes a capillary tip flow chamber configured to receive respective capillary tips and to conduct fluid past the capillary tips, and an electrode flow chamber in which an electrode is disposed and configured to conduct fluid past the electrode, the electrode flow chamber being separate from and in fluid communication with the capillary tip flow chamber. An ultraviolet (UV) light absorbance-based multiplexed capillary electrophoresis system includes a first enclosure and a second enclosure. The first enclosure covers a UV light source, and includes a slit. The second enclosure covers the first enclosure, a collimating lens, and a capillary window.

Claims (44)

1. A capillary electrophoresis system, comprising:

a console configured to house a replaceable capillary array comprising a plurality of capillaries;

a power supply disposed in the console and configured to apply a voltage across the plurality of capillaries effective for performing capillary electrophoresis on a sample in one or more capillaries of the plurality of capillaries; and

a capillary reservoir, comprising:

a fluid input port;

a capillary tip flow chamber configured to receive respective capillary tips of the plurality of capillaries and to conduct fluid past the capillary tips;

an electrode;

an electrode flow chamber in which the electrode is disposed and configured to conduct fluid past the electrode, the electrode flow chamber separate from and in fluid communication with the capillary tip flow chamber;

a waste port; and

a tube defining a fluid flow path from the fluid input port to the capillary tip flow chamber, to the electrode flow chamber, and to the waste port,

wherein a highest point of the electrode flow chamber is lower than a lowest point of the capillary tip flow chamber.

2. The capillary electrophoresis system of claim 1 , comprising a detector disposed in the console and configured to detect light transmitted from the plurality of capillaries.

The capillary electrophoresis system of claim 1 , comprising a plurality of drawers disposed in the console and configured to hold a plurality of multi-well plates, each drawer movable between a closed position inside the console and an open position outside the console at which the drawer is externally accessible.

4. The capillary electrophoresis system of claim 3 , comprising a motion control system disposed in the console and configured to move one or more multi-well plates of the plurality of multi-well plates to and/or from one or more respective drawers of the plurality of drawers.

The capillary electrophoresis system of claim 4 , wherein the console comprises an injection position at which the replaceable capillary array is received, and the motion control system is configured to move the one or more multi-well plates from the one or more respective drawers to the injection position.

6. The capillary electrophoresis system of claim 4 , wherein the motion control system comprises a tray carrier configured to support a selected multi-well plate of the plurality of multi-well plates.

The capillary electrophoresis system of claim 6 , wherein the motion control system comprises an x-drive motor configured to drive motion of the tray carrier back and forth relative to the plurality of drawers, and a z-drive motor configured to drive motion of the tray carrier up and down relative to the plurality of drawers.

8. The capillary electrophoresis system of claim 6 , wherein the tray carrier comprises one or more alignment pins configured to engage a tray holder of the selected multi-well plate.

9. The capillary electrophoresis system of claim 3 , wherein each drawer comprises one or more alignment pins configured to engage a tray holder of a selected multi-well plate of the plurality of multi-well plates.

The capillary electrophoresis system of claim 1 , wherein the capillary tips received in the capillary tip flow chamber are first capillary tips, the capillaries comprise respective second capillary tips opposite to the corresponding first capillary tips, and the system further comprises a plurality of injection electrodes communicating with the power supply and disposed proximate to the second capillary tips.

11. The capillary electrophoresis system of claim 1 , comprising a control device programmed to control an operation of the capillary electrophoresis system, the operation comprising:

controlling the system to inject the sample into the replaceable capillary array; and

controlling the power supply to, after injection of the sample, apply the voltage.

The capillary electrophoresis system of claim 11 , comprising a motion control system disposed in the console and configured to move one or more multi-well plates of a plurality of multi-well plates to and/or from an injection position of the console, wherein the operation controlled by the control device further comprises:

controlling the motion control system to, before the injecting, move a selected multi-well plate of the plurality of multi-well plates to the injection position.

13. The capillary electrophoresis system of claim 1 , comprising:

a UV light source;

a first enclosure covering the UV light source, wherein the first enclosure comprises a slit;

a collimating lens configured to be disposed between the UV light source and a capillary window of the replaceable capillary array; and

a second enclosure configured to cover the first enclosure, the collimating lens, and the capillary window.

14. The capillary electrophoresis system of claim 1 , wherein at least a portion of the tube interconnects the capillary tip flow chamber and the electrode flow chamber.

The capillary electrophoresis system of claim 1 , wherein the tube comprises a first tube portion interconnecting the fluid input port and the capillary tip flow chamber, a second tube portion interconnecting the capillary tip flow chamber and the electrode flow chamber, and a third portion interconnecting the electrode flow chamber and the waste port.

16. An ultraviolet (UV) light absorbance-based capillary electrophoresis system, comprising:

a console configured to house a replaceable capillary array comprising a plurality of capillaries and a capillary window;

a power supply disposed in the console and configured to apply a voltage across each of the plurality of capillaries effective for performing capillary electrophoresis on a sample in one or more of the capillaries;

a UV light source;

a first enclosure covering the UV light source, wherein the first enclosure comprises a slit;

a collimating lens positioned to receive UV light from the UV light source, and configured to direct collimated UV light to the capillary window; and

a second enclosure disposed in the console and covering the first enclosure and the collimating lens,

wherein the second enclosure is configured and positioned to cover the capillary window and a covered portion of the plurality of capillaries, and is configured and positioned to block airflow in the console from accessing the UV light source, the collimating lens, the capillary window, and the covered portion of the plurality of capillaries.

17. The capillary electrophoresis system of claim 16 , wherein the slit is less than 1500 microns in width.

18. The capillary electrophoresis system of claim 16 , comprising a detector disposed in the console and configured to detect light transmitted from the plurality of capillaries for acquisition of absorbance data.

19. The capillary electrophoresis system of claim 16 , comprising a fan disposed in the console and configured to direct the airflow to an exposed portion of the plurality of capillaries that is outside the second enclosure.

20. The capillary electrophoresis system of claim 16 , comprising the replaceable capillary array, wherein the replaceable capillary array is disposed in the console, and the plurality of capillaries comprises an exposed portion that is outside the second enclosure and exposed to the airflow.
