Abstract

A method and system for buoyant separation of a target constituent of a sample, the method comprising: at a process chamber, combining a volume of substrates having a first density with the sample, thereby producing a population of target-bound complexes comprising the target constituent bound to at least a portion of the volume of substrates; within the process chamber, physically separating the population of target-bound complexes from the sample based upon interaction between the volume of substrates and an applied force; aggregating the population of target-bound complexes at a collection region of the process chamber; extracting the population of target-bound complexes from the process chamber; and processing the target constituent from the population of target-bound complexes for further analysis.

Claims (34)

1. A method for buoyant separation of a set of target constituents of a sample, the method comprising:

at a single vessel, receiving:

the sample comprising the set of target constituents and a set of non-target constituents, wherein bulk fluid of the sample has a first density; and

a volume of buoyant particles having a second density, the second density less than the first density, wherein the buoyant particles are configured to bind with at least a portion of the set of target constituents thereby producing a set of target-bound complexes, wherein the volume of buoyant particles and the set of target constituents are received at the single vessel at a ratio between 1:2 and 5:1;

separating the set of target-bound complexes from the bulk fluid of the sample by enabling movement of the set of target-bound complexes to a superior region of the single vessel, thereby producing the buoyant separation; and

processing the set of non-target constituents in the single vessel.

2. The method of claim 1 , further comprising a mixing process, wherein the mixing process is configured to increase a speed at which the set of target-bound complexes moves toward the superior region.

3. The method of claim 2 , wherein the mixing process is an end-over-end mixing process.

4. The method of claim 1 , wherein each of the set of buoyant particles is a glass microbubble.

5. The method of claim 4 , wherein the second density is between 0.4 and 0.8 grams-per-centimeter-cubed.

6. The method of claim 1 , further comprising processing the set of target-bound complexes in a second vessel.

7. The method of claim 1 , further comprising processing the set of target-bound complexes in the single vessel.

8. The method of claim 7 , further comprising draining the set of non-target constituents from the single vessel prior to processing the set of target-bound complexes in the single vessel.

9. The method of claim 8 , wherein the single vessel comprises a valve configured to drain the set of non-target constituents from an outlet of the single vessel.

10. The method of claim 9 , wherein the outlet is arranged at an inferior-most region of the single vessel.

11. The method of claim 1 , wherein the ratio is between 1:1 and 2.5:1.

12. A system for buoyant separation of a set of target constituents of a sample, the system comprising:

a mixture, comprising:

a volume of buoyant particles having a first density; and

a sample comprising a set of target constituents and a set of non-target constituents, wherein bulk fluid of the sample has a second density greater than the first density;

wherein:

the volume of buoyant particles and the set of target constituents have a ratio in the mixture of between 1:2 and 5:1;

the buoyant particles are configured to bind with at least a portion of the set of target constituents, thereby producing a set of target-bound complexes;

a single vessel defining a cavity, wherein the single vessel:

receives the mixture;

enables movement of the set of target-bound complexes towards a superior region of the single vessel, thereby producing the buoyant separation, wherein the set of non-target constituents is processed in the single vessel.

13. The system of claim 12 , wherein the single vessel comprises a valve.

14. The system of claim 13 , wherein the valve is arranged at an inferior region of the single vessel.

15. The system of claim 14 , wherein the valve is configured to drain the set of non-target constituents from the inferior region of the single vessel after the set of target-bound complexes have been concentrated at the superior region of the single vessel.

16. The system of claim 12 , wherein each of the set of buoyant particles is a glass microbubble.

17. The system of claim 16 , wherein the first density is between 0.4 and 0.8 grams-per-centimeter-cubed.

18. The system of claim 12 , wherein the ratio is between 1:1 and 2.5:1.

19. The system of claim 12 , wherein the cavity is a single cavity.

20. The method of claim 1 , wherein the ratio is configured to achieve complexification of the set of target constituents to the volume of buoyant particles with a binding efficiency of at least 90%.
