Abstract

Described herein are neuroactive steroids of the Formula (I): or a pharmaceutically acceptable salt thereof, wherein A, R 1 , R 2 , R 3a , R 4a , R 4b , R 5 , R 7a , and R 7b are as defined herein. Such compounds are envisioned, in certain embodiments, to behave as GABA modulators. The present invention also provides pharmaceutical compositions comprising a compound of the present invention and methods of use and treatment, e.g., such for inducing sedation and/or anesthesia.

74 . A method of treating major depressive disorder in a human subject, comprising administering to the human subject a therapeutically effective amount of a compound having a structure:

or a pharmaceutically acceptable salt thereof.

75 . The method according to claim 74 , wherein the compound or pharmaceutically acceptable salt thereof is a citrate salt of the compound

76 . The method of claim 74 , wherein the compound or pharmaceutically acceptable salt thereof is administered in combination with another therapeutic agent.

77 . The method of claim 74 , wherein compound or pharmaceutically acceptable salt thereof is administered orally.

78 . The method of claim 77 , wherein the compound or pharmaceutically acceptable salt thereof is administered as a tablet.

79 . The method of claim 78 , wherein the tablet comprises from about 0.1% to about 50% of the compound or pharmaceutically acceptable salt thereof by weight of the tablet.

80 . The method of claim 74 , wherein the compound or pharmaceutically acceptable salt thereof is administered one to five times per day.

81 . The method of claim 80 , wherein each administration of the compound or pharmaceutically acceptable salt thereof provides from about 0.01 to about 20 mg of the compound per kilogram of body weight.

82 . The method of claim 74 , wherein the compound or pharmaceutically acceptable salt thereof is administered chronically.

83 . The method of claim 82 , wherein the compound or pharmaceutically acceptable salt thereof is administered for a period of at least 3 months.

84 . The method of claim 75 , wherein the citrate salt of the compound is administered in combination with another therapeutic agent.

85 . The method of claim 75 , wherein the citrate salt of the compound is administered is administered orally.

86 . The method of claim 85 , wherein the citrate salt of the compound is administered is administered as a tablet.

87 . The method of claim 86 , wherein the tablet comprises from about 0.1% to about 50% of the compound or pharmaceutically acceptable salt thereof by weight of the tablet.

88 . The method of claim 86 , wherein the citrate salt of the compound is administered is administered one to five times per day.

89 . The method of claim 88 , wherein each administration of the citrate salt of the compound provides from about 0.01 to about 20 mg of the compound per kilogram of body weight.

90 . The method of claim 75 , wherein the citrate salt of the compound is administered chronically.

91 . The method of claim 90 , wherein the citrate salt is administered for a period of at least 3 months.

92 . A method of treating major depressive disorder in a human subject, comprising administering to the human subject a therapeutically effective amount of a pharmaceutical composition comprising compound having a structure:

or a pharmaceutically acceptable salt thereof.

93 . The method of claim 92 , wherein the compound or pharmaceutically acceptable salt of the compound is a citrate salt of the compound

94 . The method of claim 92 , wherein the pharmaceutical composition is administered orally.

95 . The method of claim 94 , wherein the pharmaceutical composition is a tablet.

96 . The method of claim 95 , wherein the tablet comprises from about 0.1% to about 50% of the compound or pharmaceutically acceptable salt thereof by weight of the tablet.

97 . The method of claim 92 , wherein the pharmaceutical composition is administered one to five times per day.

98 . The method of claim 97 , wherein each administration of the pharmaceutical composition provides from about 0.01 to about 20 mg of the compound per kilogram of body weight.

99 . The method of claim 93 , wherein the pharmaceutical composition is administered orally.

100 . The method of claim 99 , wherein the pharmaceutical composition is a tablet.

101 . The method of claim 100 , wherein the tablet comprises from about 0.1% to about 50% of the citrate salt of the compound by weight of the tablet.

102 . The method of claim 93 , wherein the pharmaceutical composition is administered one to five times per day.

103 . The method of claim 102 , wherein each administration of the pharmaceutical composition provides from about 0.01 to about 20 mg of the compound per kilogram of body weight.
