Abstract

Cyclic GMP-AMP synthase (cGAS) enzymes have been engineered to produce polypeptides having increased cGAS activity in the cyclization of modified nucleoside triphosphates, including thiolated or fluorinated nucleoside triphosphates, and polynucleotides encoding these enzymes, as well as vectors and host cells comprising these polynucleotides and polypeptides. Methods for producing cGAS enzymes, compositions comprising the cGAS enzymes, and methods of using the engineered cGAS enzymes are useful for the production of pharmaceutical compounds.

Claims (18)

1. An engineered cGAS enzyme comprising a polypeptide sequence having at least 95%, sequence identity to SEQ ID NO: 2 with the substitutions G163N, Y201F, C389S, R255K and L334I, wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 2, and wherein the engineered cGAS enzyme exhibits improved production of a cyclic dinucleotide.

2. The engineered cGAS enzyme of claim 1 further comprising at least one substitution at one or more positions selected from 131, 135, 139, 160, 161, 164, 204, 286, and 392, wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 318.

The engineered cGAS enzyme of claim 1 , further comprising at least one substitution at one or more positions selected from 127, 136, 147, 164, 169, 280, 299, 303, 364, 367, 382, 408, 412, and 413, wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 556.

4. The engineered cGAS enzyme of claim 1 , further comprising at least one substitution at one or more positions selected from 127, 147, 164, 169, 189, 210, 367, 408, 413, and 449, wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 556.

5. The engineered cGAS enzyme of claim 1 further comprising at least one substitution at one or more positions selected from 8, 127, 132, 136, 147, 150, 303, 312, and 412, wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 556.

The engineered cGAS enzyme of claim 1 further comprising at least one substitution at one or more positions selected from 44, 113, 206, 210, 212 253, 265, 376, 379, 382, 383, 393, 408, 413, 466 and 474, wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 566.

7. The engineered cGAS enzyme of claim 1 further comprising at least one substitution at one or more positions selected from 210, 212, 272, 299, 376, 382, 383, 388, 408, 413 and 466, wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 666.

8. The engineered cGAS enzyme of claim 1 , wherein said engineered cGAS enzyme comprises a variant engineered cGAS enzyme set forth in SEQ ID NOs: 318, 556, 566, and/or 666.

The engineered cGAS enzyme of claim 1 , wherein said engineered cGAS enzyme comprises a polypeptide sequence that is at least 95%, 96%, 97%, 98%, or 99% identical to the sequence of at least one SEQ ID NO: selected from the group consisting of: 318, 336, 354, 360, 436, 484, 540, 542, 544, 546, 548, 550, 552, 554, 556, 558, 560, 562, 564, 566, 568, 570, 572, 574, 576, 578, 580, 582, 584, 586, 588, 590, 592, 594, 596, 598, 600, 602, 604, 606, 608, 610, 612, 614, 616, 618, 620, 622, 624, 626, 628, 630, 632, 634, 636, 638, 640, 642, 644, 646, 648, 650, 652, 654, 656, 658, 660, 662, 664, 666, 668, 670, 672, 674, 676, 678, 680, 682, 684, 686, 688, 690, 692, 694, 696, 698, 700, 702, 704, 706, 708, 710, 712, 714, 716, 718, 720, 722, 724, 726, 728, 730, 732, 734, 736, 738, 740, 742, 744, 746, 748, 750, 752, 754, 756, 758, 760, 762, 764, 766, 768, 770, 772, 774, 776, 778, 780, 782, 784, 786, 788, 790, 792, 794, 796, 798, 800, 802, 804, 806, 808, 810, and 812.

11. The engineered cGAS enzyme of claim 1 , wherein said engineered cGAS enzyme comprises at least one improved property compared to the wild-type Haliaeetus leucocephalus cGAS enzyme.

12. The engineered cGAS enzyme of claim 11 , wherein said improved property comprises improved activity on a substrate.

13. The engineered cGAS enzyme of claim 12 , wherein said substrate comprises Sp-3′Fluoro-3′-deoxyguanosine-5′-(1-thio)-triphosphate (F-thioGTP or compound (2)) and/or Sp-2′Fluoro-ara-adenosine-5′-1-thio-triphosphate (F-thioATP or compound (3)).

14. The engineered cGAS enzyme of claim 11 , wherein said improved property comprises improved production of compound (1)

15. The engineered cGAS enzyme of claim 11 , wherein said improved property comprises increased activity, increased substrate tolerance, and/or increased stability.

16. The engineered cGAS enzyme of claim 1 , wherein said engineered cGAS enzyme is purified.

17. A composition comprising at least one engineered cGAS enzyme of claim 1 .

18. The engineered cGAS enzyme of claim 1 comprising at least one substitution at one or more positions selected from 135, 149, 159, 164, 181, 190, 204, 262, 286, and 464 wherein the amino acid positions of said polypeptide sequence are numbered with reference to SEQ ID NO: 34.
