Abstract

The present disclosure relates to anti-IL-27 antibodies, and antigen-binding portions thereof. The disclosure also relates to methods for treating or ameliorating one or more symptoms of a disease, such as cancer, by administering the antibodies or antigen-binding portion thereof. The disclosure also relates to methods for detecting IL-27 in, for example, a subject or a sample.

Claims (14)

1 . A method of treating an interleukin 27 (IL-27)-expressing cancer in a subject, the method comprising administering to the subject an effective amount of an isolated monoclonal antibody that specifically binds to and antagonizes human IL-27, or antigen binding portion thereof, wherein the antibody or antigen binding portion thereof comprises:

heavy chain CDR1, CDR2 and CDR3 sequences comprising SEQ ID NOs: 161, 162 and 163, respectively, and light chain CDR1, CDR2 and CDR3 sequences comprising SEQ ID NOs: 169, 170 and 171, respectively; or

heavy chain CDR1, CDR2 and CDR3 sequences comprising SEQ ID NOs: 164, 165, and 166, respectively, and light chain CDR1, CDR2 and CDR3 sequences comprising SEQ ID NOs: 172, 173, and 174, respectively.

The method of claim 1 , wherein the antibody or antigen binding portion thereof comprises the heavy chain CDR1, CDR2 and CDR3 sequences comprising SEQ ID NOs: 161, 162 and 163, respectively, and the light chain CDR1, CDR2 and CDR3 sequences comprising SEQ ID NOs: 169, 170 and 171, respectively.

The method of claim 2 , wherein the IL-27-expressing cancer is selected from the group consisting of renal cell carcinoma (RCC), hepatocellular carcinoma (HCC), lung cancer, Hodgkin's disease, sarcoma, testicular cancer, ovarian cancer, pancreas cancer, breast cancer, melanoma, head and neck cancer, colorectal cancer, bladder cancer, endometrial cancer, prostate cancer, thyroid cancer, gastroesophageal cancer, gastric cancer, brain cancer, lymphoma, and leukemia.

4 . The method of claim 2 , wherein the isolated monoclonal antibody, or antigen binding portion thereof, is administered in combination with one or more additional therapeutic agents or procedures.

5 . The method of claim 4 , wherein the one or more additional therapeutic agents or procedures is selected from the group consisting of a chemotherapy; a cytokine; a surgical procedure; a radiation procedure; and a combination thereof.

6 . The method of claim 2 , wherein the isolated monoclonal antibody or antigen binding portion thereof comprises a heavy chain variable region comprising SEQ ID NO: 167 and a light chain variable region comprising SEQ ID NO: 175.

The method of claim 2 , wherein the isolated monoclonal antibody or antigen binding portion thereof comprises a heavy chain comprising SEQ ID NO: 181 and a light chain comprising SEQ ID NO: 179, or the isolated monoclonal antibody or antigen binding portion thereof comprises a heavy chain comprising SEQ ID NO: 177 and a light chain comprising SEQ ID NO: 179.

8 . The method of claim 4 , wherein the one or more additional therapeutic agents is selected from the group consisting of: PDR001, nivolumab, pembrolizumab, pidilizumab, MEDI0680, REGN2810, TSR-042, PF-06801591, and AMP-224.

9 . The method of claim 4 , wherein the one or more additional therapeutic agents is selected from the group consisting of: FAZ053, Atezolizumab, Avelumab, Durvalumab, and BMS-936559.

The method of claim 4 , wherein the one or more additional therapeutic is selected from the group agents consisting of sunitinib, cabozantinib, axitinib, lenvatinib, everolimus, bevacizumab, epacadostat, NKTR-214, tivozanib, abexinostat, pilimumab, tremelimumab, pazopanib, sorafenib, temsirolimus, ramucirumab, niraparib, savolitinib, vorolanib, regorafenib, donafenib, camrelizumab, pexastimogene, devacirepvec, ramucirumab, apatinib, encapsulated doxorubicin, tivantinib, ADI-PEG 20, binimetinib, apatinib mesylate, nintedanib, lirilumab, nivolumab, pembrolizumab, atezolizumab, avelumab, durvalumab, cemiplimab, tislelizumab, and spartalizumab.

11 . The method of claim 4 , wherein the one or more additional therapeutic agents is a TIM-3 inhibitor, wherein the TIM-3 inhibitor is MGB453 or TSR-022.

12 . The method of claim 4 , wherein the one or more additional therapeutic agents is a LAG-3 inhibitor, wherein the LAG-3 inhibitor is selected from the groups consisting of LAAG525, BMS-986016, and TSR-033.
