Abstract

The present disclosure relates to the field of synthesizing substantially pure varenicline and its intermediates. It also relates to the pharmaceutical compositions comprising varenicline and the method of use of these pharmaceutical compositions for smoking cessation.

Claims (15)

1. A pharmaceutical composition in the form of a tablet, comprising varenicline tartrate and means for controlling the level of N-nitroso-varenicline (7,8,9,10-tetrahydro-8-nitroso-6,10-Methano-6H-pyrazino[2,3-h][3] benzazepine) impurity to less than 50 ppm per tablet as measured by LC-ESI-HRMS (U.S. FDA Method), wherein the varenicline tartrate is dispersed in a maltodextrin matrix.

2. A pharmaceutical composition, comprising varenicline tartrate maltodextrin premix having less than 50 ppm N-nitroso-varenicline (7,8,9,10-tetrahydro-8-nitroso-6,10-Methano-6H-pyrazino[2,3-h][3] benzazepine) impurity as measured by LCMS.

3. The pharmaceutical composition of claim 2 , comprising generally spherical agglomerations of varenicline tartrate and maltodextrin.

4. The pharmaceutical composition of claim 2 , wherein the varenicline tartrate is present in an amount of about 0.85 mg to about 1.7 mg, the maltodextrin is present in an amount of about 5 mg to about 50 mg, and wherein the composition further comprises:

a. about 5 mg to about 15 mg croscarmellose sodium;

b. about 100 mg to about 200 mg microcrystalline cellulose; and

c. about 0.5 mg to about 2 mg stearic acid,

wherein the composition is formulated into a tablet and comprises less than 15 ppm N-nitroso-varenicline impurity per tablet as measured by LC-ESI-HRMS (U.S. FDA Method).

5. The pharmaceutical composition of claim 2 , wherein the composition is in the form of a tablet, comprises about 0.85 mg to about 1.7 mg varenicline tartrate, and comprises less than 25 ppm N-nitroso-varenicline impurity per tablet as measured by LC-ESI-HRMS (U.S. FDA Method).

6. The pharmaceutical composition of claim 2 , wherein the varenicline tartrate is approximately uniformly dispersed in the maltodextrin matrix.

7. The pharmaceutical composition of claim 2 , wherein the premix is spray-dried.

8. The pharmaceutical composition of claim 2 , wherein the composition is formulated into a tablet comprising varenicline tartrate, microcrystalline cellulose and croscarmellose sodium having less than 20 ppm N-nitroso-varenicline impurity as measured by LC-ESI-HRMS (U.S. FDA Method).

9. The pharmaceutical composition of claim 1 , wherein the tablet comprises about 1.7 mg of the varenicline tartrate, or about 0.85 mg of the varenicline tartrate, and has less than 25 ppm of the N-nitroso-varenicline impurity per tablet.

10. The pharmaceutical composition of claim 1 , wherein the tablet comprises about 1.7 mg of the varenicline tartrate, or about 0.85 mg of the varenicline tartrate, and has less than 19 ppm of the N-nitroso-varenicline impurity per tablet.

11. The pharmaceutical composition of claim 1 , wherein the tablet comprises about 1.7 mg of the varenicline tartrate, or about 0.85 mg of the varenicline tartrate, and has less than 5 ppm of the N-nitroso-varenicline impurity per tablet.
