Abstract

A bioactive suspension derived from freshly disaggregated tissue is provided, as well as related methods of formulation and use. The bioactive suspension may comprise a cell-free supernate derived from epidermal and dermal tissue that has been enzymatically and mechanically disaggregated, then separated, and which may contain tissue regeneration factors known to speed healing. The bioactive suspension may further comprise genetically-modified treatment cells, wild type cells, or both, and may be combined with one or more scaffolding elements to form a bioactive suspension combination product suitable for treatment of a cutaneous defect. Synthetic bioactive suspensions and bioactive suspension combination products are also provided.

Claims (16)

1 . A method of treatment, comprising the steps of:

a. obtaining a tissue sample;

b. disaggregating the tissue sample to produce a disaggregated tissue sample;

c. adding an effective amount of buffer to the disaggregated tissue sample to produce a cell suspension, wherein the cell suspension contains cells;

d. allowing the cell suspension to rest an effective amount of time;

e. separating out the cells from the cell suspension by at least one cell separation procedure to produce a bioactive suspension, wherein the bioactive suspension contains at least one tissue regeneration factor; and

f. applying the bioactive suspension to at least one treatment site.

2 . The method of claim 1 further comprising, prior to the step of applying the bioactive suspension to at least one treatment site, the step of:

adding the bioactive suspension to at least one scaffolding element.

3 . The method of claim 1 further comprising, prior to the step of applying the bioactive suspension to at least one treatment site, the step of:

adding at least one population of treatment cells.

4 . The method of claim 1 further comprising, prior to the step of applying the bioactive suspension to at least one treatment site, the step of:

adding at least one efficacious element.

5 . The method of claim 1 further comprising, prior to the step of applying the bioactive suspension to at least one treatment site, the steps of:

freezing the bioactive suspension; and

thawing the bioactive suspension.
