Abstract

Oral pharmaceutical compositions of gamma-hydroxybutyrate (GHB) suitable for concomitant administration with a dose of divalproex sodium (DVP) without materially altering the dosage amount of either drug are provided. Also provided are therapeutic uses of the compositions for the treatment of one or more symptoms of narcolepsy.

Claims (28)

1 . A composition for the treatment of narcolepsy or a symptom thereof, wherein the composition comprises gamma-hydroxybutyrate, and wherein administration of a single 6 g dose of the composition with divalproex sodium ER at steady-state results in an approximate 18% increase in AUC, with a 90% confidence interval (CI) within a 80-125% bioequivalence range, relative to administration of a single 6 g dose of the composition without divalproex sodium ER.

2 . The composition of claim 1 , wherein the composition is suitable for administration only once daily.

3 . The composition of claim 1 , wherein the composition is effective to induce sleep in the human subject for at least six hours.

4 . The composition of claim 1 , wherein the composition is a particulate formulation that forms a suspension when mixed with water.

5 . The composition of claim 1 , further comprising an immediate release portion and a modified release portion, wherein each of the immediate release portion and the modified release portion comprises gamma-hydroxybutyrate.

6 . The composition of claim 5 , wherein a ratio of gamma-hydroxybutyrate in the immediate release portion and the modified release portion is from 10/90 to 65/35.

7 . The composition of claim 1 , wherein the gamma-hydroxybutyrate comprises 3.0 g to 12.0 g of sodium oxybate.

A composition for the treatment of narcolepsy or a symptom thereof, wherein the composition comprises gamma-hydroxybutyrate, and wherein administration of a single 6 g dose of the composition with divalproex sodium ER at steady-state results in a C max bioequivalent to administration of a single 6 g dose of the composition without divalproex sodium ER.

9 . The composition of claim 8 , wherein the composition is suitable for administration only once daily.

10 . The composition of claim 8 , wherein the composition is effective to induce sleep in the human subject for at least six hours.

11 . The composition of claim 8 , wherein the composition is a particulate formulation that forms a suspension when mixed with water.

12 . The composition of claim 8 , further comprising an immediate release portion and a modified release portion, wherein each of the immediate release portion and the modified release portion comprises gamma-hydroxybutyrate.

13 . The composition of claim 12 , wherein a ratio of gamma-hydroxybutyrate in the immediate release portion and the modified release portion is from 10/90 to 65/35.

14 . The composition of claim 8 , wherein the gamma-hydroxybutyrate comprises 3.0 g to 12.0 g of sodium oxybate.

15 . A composition for the treatment of a human disorder treatable with gamma-hydroxybutyrate, wherein composition comprises a daily dose of gamma-hydroxybutyrate in an amount equivalent to from 0.5 g to 12.0 g of sodium oxybate, and wherein administration of a dose of the composition with divalproex sodium ER at steady-state results in an AUC bioequivalent to administration of a dose of the composition without divalproex sodium ER.

16 . The composition of claim 15 , wherein the composition is suitable for administration only once daily.

17 . The composition of claim 15 , wherein the composition is effective to induce sleep in the human subject for at least six hours.

18 . The composition of claim 15 , wherein the composition is a particulate formulation that forms a suspension when mixed with water.

19 . The composition of claim 15 , further comprising an immediate release portion and a modified release portion, wherein each of the immediate release portion and the modified release portion comprises gamma-hydroxybutyrate.

20 . The composition of claim 19 , wherein a ratio of gamma-hydroxybutyrate in the immediate release portion and the modified release portion is from 10/90 to 65/35.

21 . The composition of claim 15 , wherein the gamma-hydroxybutyrate comprises 3.0 g to 12.0 g of sodium oxybate.

22 . A composition for the treatment of a human disorder treatable with gamma-hydroxybutyrate, wherein the composition comprises gamma-hydroxybutyrate, and wherein administration of a single 6 g dose of the composition with divalproex sodium ER at steady-state does not result in a pharmacokinetic interaction in adults.

23 . The composition of claim 22 , wherein the composition is suitable for administration only once daily.

24 . The composition of claim 22 , wherein the composition is effective to induce sleep in the human subject for at least six hours.

25 . The composition of claim 22 , wherein the composition is a particulate formulation that forms a suspension when mixed with water.

26 . The composition of claim 22 , further comprising an immediate release portion and a modified release portion, wherein each of the immediate release portion and the modified release portion comprises gamma-hydroxybutyrate.

27 . The composition of claim 26 , wherein a ratio of gamma-hydroxybutyrate in the immediate release portion and the modified release portion is from 10/90 to 65/35.

28 . The composition of claim 22 , wherein the gamma-hydroxybutyrate comprises 3.0 g to 12.0 g of sodium oxybate.
