Bupropion as a modulator of drug activity
Dosage forms, drug delivery systems, and methods related to sustained release of dextromethorphan or improved therapeutic effects are disclosed. Typically, bupropion or a related compound is orally administered to a human being to be treated with, or being treated with, dextromethorphan.
1. A method of treating depression while reducing the risk of an adverse event associated with treatment with bupropion, comprising orally administering a daily dose of bupropion that is about 200 mg to about 250 mg of bupropion hydrochloride, or a molar equivalent amount of bupropion in another salt form or the free base form, and a daily dose of dextromethorphan that is about 80 mg to about 100 mg of dextromethorphan hydrobromide, or a molar equivalent amount of dextromethorphan in another salt form or the free base form, to a human being in need thereof for at least 8 consecutive days, wherein the daily dose of the bupropion and the dextromethorphan achieves the same or an improved therapeutic effect in treating depression as compared to administering a higher daily dose of bupropion alone, thereby reducing the risk of the adverse event associated with treatment with bupropion.
2. The method of claim 1 , wherein about 105 mg of bupropion hydrochloride is orally administered twice a day to the human being.
3. The method of claim 2 , wherein about 45 mg of dextromethorphan hydrobromide is orally administered twice a day to the human being.
4. The method of claim 1 , wherein the adverse event is headache.
5. The method of claim 1 , wherein the adverse event is insomnia.
6. The method of claim 1 , wherein the adverse event is irritability.
7. The method of claim 1 , wherein the adverse event is nausea.
8. The method of claim 1 , wherein the adverse event is anxiety.
9. The method of claim 1 , wherein the adverse event is weight gain.
10. The method of claim 1 , wherein the adverse event is seizure.
11. The method of claim 1 , wherein the human being is suffering from major depressive disorder.
12. The method of claim 1 , wherein the human being is suffering from treatment-resistant major depressive disorder.
13. A method of treating depression while reducing the risk of an adverse event associated with treatment with bupropion, comprising orally administering a combination of bupropion that is about 70 mg to about 95 mg of the bupropion free base, or a molar equivalent amount of bupropion in a salt form, and dextromethorphan that is about 30 mg to about 42 mg of the dextromethorphan free base, or a molar equivalent amount of dextromethorphan in a salt form, to a human being in need thereof once a day or twice a day for at least 8 consecutive days, wherein administering the combination of the bupropion and the dextromethorphan achieves the same or an improved therapeutic effect in treating depression as compared to administering a higher molar equivalent amount of bupropion alone, thereby reducing the risk of the adverse event associated with treatment with bupropion.
14. The method of claim 13 , wherein the adverse event is headache.
15. The method of claim 13 , wherein the adverse event is insomnia.
16. The method of claim 13 , wherein the adverse event is irritability.
17. The method of claim 13 , wherein the adverse event is nausea.
18. The method of claim 13 , wherein the adverse event is anxiety.
19. The method of claim 13 , wherein the adverse event is weight gain.
20. The method of claim 13 , wherein the adverse event is seizure.
21. The method of claim 13 , wherein the human being is suffering from major depressive disorder.
22. The method of claim 13 , wherein the human being is suffering from treatment-resistant major depressive disorder.
