Abstract

This invention provides novel injectable compositions comprising botulinum toxin that may be administered to a subject for various therapeutic, aesthetic and/or cosmetic purposes. The injectable compositions contemplated by the invention exhibit one or more advantages over conventional botulinum toxin formulations, including reduced antigenicity, a reduced tendency to undergo unwanted localized diffusion following injection, increased duration of clinical efficacy or enhanced potency relative, faster onset of clinical efficacy, and/or improved stability.

Claims (43)

1 . A method of administering botulinum toxin to achieve a therapeutic or cosmetic effect to an individual in need thereof, the method comprising

injecting an effective amount of a composition into the individual to achieve a therapeutic or cosmetic effect,

wherein the composition comprises a positively charged carrier with the amino acid sequence RKKRRQRRR-(G)p-(K)n-(G)q-RKKRRQRRR, wherein n is selected from 10 to 40, p is selected from 0 to 5, and q is selected from 0 to 5,

a botulinum toxin complex, reduced botulinum toxin complex, or botulinum toxin, and

wherein the positively charged carrier is non-covalently associated with the botulinum toxin complex, reduced botulinum toxin complex, or botulinum toxin.

2 . The method according to claim 1 , wherein the botulinum toxin complex, reduced botulinum toxin complex or botulinum toxin is obtained from serotypes A of C. botulinum.

6 . The method according to claim 1 , wherein the positively charged carrier stabilizes botulinum toxin against degradation.

7 . The method according to claim 1 , wherein the positively charged carrier reduces local diffusion of the botulinum toxin following injection.

8 . The method according to claim 1 , wherein the composition has a reduced tendency to induce antibody generation following injection into the subject's body compared to BOTOX®.

9 . The method according to claim 1 , wherein the cosmetic effect is a reduction of wrinkles.

The method according to claim 1 , wherein the therapeutic effect is reduction of a symptom associated with a disorder selected from the group consisting of hemifacial spasm, adult onset spasmodic torticollis, anal fissure, blepharospasm, cerebral palsy, headaches, strabismus, temporomandibular joint disorder, neurologic pain, overactive bladder, rhinitis, sinusitis, acne, dystonia, dystonic contractions, hyperhidrosis, and hypersecretion of a gland controlled by the cholinergic nervous system.

11 . A method of preparing a sterile injectable composition according to claim 19 , the method comprising

providing an effective amount of a botulinum toxin complex, a reduced botulinum toxin complex, or a botulinum toxin;

providing a positively charged carrier with the amino acid sequence RKKRRQRRR-(G)p-(K)n-(G)q-RKKRRQRRR, wherein n is selected from 10 to 40, p is selected from 0 to 5, and q is selected from 0 to 5;

combining the botulinum toxin complex, a reduced botulinum toxin complex, or botulinum toxin with the positively charged carrier and a pharmaceutically acceptable diluent.

12 . The method according to claim 11 , wherein the botulinum toxin complex, reduced botulinum toxin complex, or botulinum toxin is obtained from serotype A of C. botulinum.

16 . The method according to claim 1 , wherein the positively charged carrier stabilizes botulinum toxin against degradation.

17 . The method according to claim 1 , wherein the positively charged carrier reduces local diffusion of botulinum toxin following injection.

19 . A sterile injectable composition comprising

an effective amount of a botulinum toxin complex, a reduced botulinum toxin complex, or a botulinum toxin; and

a positively charged carrier with the amino acid sequence RKKRRQRRR-(G)p-(K)n-(G)q-RKKRRQRRR, wherein n is selected from 10 to 40, p is selected from 0 to 5, and q is selected from 0 to 5;

wherein the botulinum toxin complex, reduced botulinum toxin complex, or botulinum toxin non-covalently associates with the positively charged carrier.

20 . A syringe comprising

an effective amount of a botulinum toxin complex, a reduced botulinum toxin complex, or a botulinum toxin; and

a positively charged carrier with the amino acid sequence RKKRRQRRR-(G)p-(K)n-(G)q-RKKRRQRRR, wherein n is selected from 10 to 40, p is selected from 0 to 5, and q is selected from 0 to 5;

wherein the botulinum toxin complex, reduced botulinum toxin complex, or botulinum toxin non-covalently associates with the positively charged carrier.

21 . A reconstituted botulinum toxin composition comprising

an effective amount of a botulinum toxin complex, a reduced botulinum toxin complex, or a botulinum toxin; and

a positively charged carrier with the amino acid sequence RKKRRQRRR-(G)p-(K)n-(G)q-RKKRRQRRR, wherein n is selected from 10 to 40, p is selected from 0 to 5, and q is selected from 0 to 5; and

a pharmaceutically acceptable aqueous liquid diluent,

wherein the botulinum toxin complex, reduced botulinum toxin complex, or botulinum toxin non-covalently associates with the positively charged carrier,

and wherein the reconstituted botulinum toxin composition has a viscosity in the range of about 0.4 to about 2.0 cP.

22 . The method according to claim 1 , wherein p is 0 or 1 and q is 0 or 1.

23 . The method according to claim 1 , wherein p and q are 0.

24 . The method according to claim 1 , wherein p and q are 1.

25 . The method according to claim 1 , wherein n is 15 to 30.
