Abstract

A method for providing a passivation layer or pH protective coating on a substrate surface by PECVD is provided, the method comprising generating a plasma from a gaseous reactant comprising polymerizing gases. The lubricity, passivation, pH protective, hydrophobicity, and/or barrier properties of the passivation layer or pH protective coating are set by setting the ratio of the O 2 to the organosilicon precursor in the precursor feed, and/or by setting the electric power used for generating the plasma. In particular, a passivation layer or pH protective coating made by the method is provided. Pharmaceutical packages coated by the method and the use of such packages protecting composition contained in the vessel against mechanical and/or chemical effects of the surface of the package without a passivation layer or pH protective coating material are also provided.

Claims (28)

1. A prefilled syringe comprising:

a syringe barrel having a lumen defined at least in part by a thermoplastic wall having an interior surface facing the lumen and an outer surface;

a lubricity coating of Si w O x C y , in which w is 1, x is from about 0.5 to about 2.4, and y is from about 0.6 to about 3, the lubricity coating having an inner surface facing the lumen and an outer surface facing the interior surface of the thermoplastic wall, the lubricity coating having a lower frictional resistance than an uncoated surface of the thermoplastic wall;

a fluid composition contained in the lumen, the fluid composition comprising a botulinum toxin and having a pH between 4 and 10,

a plunger tip positioned in the lumen and being slidable along the interior surface of the wall; and

a cap or needle shield at a distal end of the syringe barrel;

wherein the shelf life of the prefilled syringe is more than six months at a storage temperature of 4° C.

2. The prefilled syringe of claim 1 , wherein the lubricity coating is less than 1000 nm thick.

3. The prefilled syringe of claim 1 , wherein the lubricity coating is formed by PECVD.

4. The prefilled syringe of claim 1 , wherein the lubricity coating is formed by applying a liquid organosiloxane to the interior surface and crosslinking the liquid organosiloxane.

5. The prefilled syringe of claim 1 , wherein the wall of the syringe barrel is COC (cyclic olefin copolymer) or COP (cyclic olefin polymer).

6. The prefilled syringe of claim 1 , wherein the fluid comprising a botulinum toxin is an aqueous botulinum toxin formulation.

7. The prefilled syringe of claim 1 , in which a plunger initiation force F i of the syringe is less than 11 N, a plunger maintenance force F m of the syringe is less than 11 N, or both.

8. The prefilled syringe of claim 1 , in which the syringe is a cartridge.

9. The prefilled syringe of claim 1 , wherein the lubricity coating comprises a crosslinked organosiloxane.

10. The prefilled syringe of claim 9 , wherein the lubricity coating comprises a crosslinked product of a linear siloxane having the following formula:

in which each R is an alkyl group and n is two or greater.

11. The prefilled syringe of claim 9 , wherein the lubricity coating comprises a crosslinked product of a cyclic siloxane having the following formula:

in which each R is an alkyl group and a is from 3 to 10.

12. The prefilled syringe of claim 1 , wherein the botulinum toxin is selected from the following: Abobotulinumtoxin A, Onabotulinumtoxin A, and Incobotulinumtoxin A.

13. The prefilled syringe of claim 12 , wherein the botulinum toxin is Abobotulinumtoxin A.

14. The prefilled syringe of claim 12 , wherein the botulinum toxin is Onabotulinumtoxin A.

15. The prefilled syringe of claim 12 , wherein the botulinum toxin is Incobotulinumtoxin A.

16. The prefilled syringe of claim 1 , in which the lubricity coating is effective to reduce the plunger sliding force or breakout force at least 25 percent relative to an uncoated syringe barrel.

17. The prefilled syringe of claim 16 , in which the lubricity coating is effective to reduce the plunger sliding force or breakout force at least 45 percent relative to an uncoated syringe barrel.

18. The prefilled syringe of claim 1 , in which at least a portion of the interior surface of the wall comprises a barrier coating effective to reduce the ingress of oxygen through the wall compared to an uncoated wall.

19. The prefilled syringe of claim 18 , wherein the barrier coating comprises SiO x , wherein x is from 1.5 to 2.9.

The prefilled syringe of claim 18 , wherein the barrier coating has an inner surface facing the lumen and an outer surface facing the interior surface of the thermoplastic wall and the lubricity coating has an inner surface facing the lumen and outer surface facing the inner surface of the barrier coating.
