Abstract

Provided herein are methods for the production of native and reconstituted hyaluronan (HA) complexes containing pentraxin-3 (PTX3) and heavy chain 1 (HC1) of inter alpha inhibitor (IαI). Compositions containing the complexes and therapeutic methods using the complexes are provided. Combinations and kits for use in practicing the methods also are provided.

149 . A method of treating a cancer in an individual in need thereof, the method comprising administering a therapeutically effective amount of a Heavy Chain-Hyaluronan/Pentraxin 3 (HC-HA/PTX3) complex to the individual, thereby treating the cancer in the individual.

150 . The method of claim 149 , wherein the Hyaluronan (HA) of the HC-HA/PTX3 complex comprises high molecular weight (HMW) HA.

151 . The method of claim 149 , wherein the HC-HA/PTX3 complex comprises native HC-HA/PTX3 complex (nHC-HA/PTX3).

152 . The method of claim 151 , wherein the nHC-HA/PTX3 complex is isolated from fetal tissue that is frozen or previously frozen.

153 . The method of claim 152 , wherein the fetal tissue comprises amniotic membrane or umbilical cord extract.

154 . The method of claim 152 , wherein the nHC-HA/PTX3 complex is isolated from amniotic membrane extract.

155 . The method of claim 149 , wherein the HC-HA/PTX3 complex comprises reconstituted HC-HA/PTX3 (rcHC-HA/PTX3).

156 . The method of claim 155 , wherein the rcHC-HA/PTX3 comprises hyaluronan (HA), heavy chain 1 (HC1), and pentraxin 3 (PTX3).

157 . The method of claim 149 , wherein the individual is a human.

158 . The method of claim 149 , wherein the treating comprises inhibiting the growth of cancer cells.

159 . The method of claim 149 , wherein the treating comprises killing cancer cells.

160 . The method of claim 149 , wherein the cancer comprises prostate cancer, colorectal cancer, liver cancer, kidney cancer, bladder cancer, brain cancer, ovarian cancer, lung cancer, melanoma, breast cancer, thyroid cancer, pancreatic cancer, stomach cancer, lung cancer, leukemia, lymphoma, sarcomas, gastric cancer, bone cancer, oral cancer, eye cancer, or carcinoma.

161 . The method of claim 149 , wherein the cancer is pancreatic cancer.

162 . The method of claim 149 , wherein the HC-HA/PTX3 complex is formulated for administration by injection.

163 . The method of claim 149 , wherein the HC-HA/PTX3 complex is formulated as a powder.

164 . The method of claim 149 , wherein the HC-HA/PTX3 complex is reconstituted with a pharmaceutically acceptable excipient prior to injection.

165 . The method of claim 164 , wherein the pharmaceutically acceptable excipient is selected from the group consisting of: a natural polymer, an adjuvant, a preservative, an agent for delaying absorption, a filler, a binder, an adsorbent, a buffer, and/or a combination thereof.
