Abstract

An apparatus includes a reaction vessels coupled to an electronic sensor for monitoring a reaction product in the reaction vessel; a fluidics system for sequentially delivering a plurality of reagents to the reaction vessel, the fluidics system including a plurality of reagent reservoirs in fluidic communication via a plurality of flow paths with a fluidics circuit and to a common passage in fluidic communication between the fluidics circuit and the reaction vessel, a solution reservoir in fluidic communication with the common passage via a branch passage connected with the common passage at a junction between the fluidics circuit and the reaction vessel; and an electrode in contact with a solution within the branch passage, the electrode being in electrical communication with the reaction vessel through fluid extending from the branch passage and through the common passage, the electronic sensor generating an output signal depending on a voltage of the electrode.

Claims (29)

1 . An apparatus comprising:

a reaction vessel coupled to an electronic sensor for monitoring a reaction product in the reaction vessel, wherein the reaction vessel includes a microwell array integrated with a sensor array;

a fluidics system for sequentially delivering a plurality of reagents to the reaction vessel, the fluidics system including a plurality of reagent reservoirs in fluidic communication via a plurality of flow paths with a fluidics circuit and to a common passage in fluidic communication between the fluidics circuit and the reaction vessel, a solution reservoir containing a solution in fluidic communication with the common passage via a branch passage connected with the common passage at a junction between the fluidics circuit and the reaction vessel, and a valve disposed between the solution reservoir and the junction; and

an electrode in contact with the solution within the branch passage, the electrode being disposed between the valve and the junction, the electrode being in electrical communication with the reaction vessel through fluid extending from the branch passage and through the common passage, wherein the electrode avoids direct contact with the plurality of reagents and uses the conductive solution for electrical contact, the electronic sensor generating an output signal depending on a voltage of the electrode.

2 . The apparatus of claim 1 , wherein the electrode is in contact with the solution from the solution reservoir and does not contact any of the plurality of reagents.

3 . The apparatus of claim 1 , wherein the electrode is configured to provide a reference voltage to the electronic sensor.

The apparatus of claim 1 , wherein the reaction vessel includes a well of an array of wells disposed on an array of chemically sensitive field-effect transistor sensors, the electronic sensor comprising a chemically sensitive field-effect transistor sensor of the array of chemically sensitive field-effect transistor sensors.

5 . The apparatus of claim 4 , wherein the fluidics system comprises a flow cell in fluid communication with the wells and configured to deliver reagents of the plurality of reagents to each well of the array of wells at substantially the same flow rate.

6 . The apparatus of claim 1 , wherein substantially none of the plurality of reagents contacts the electrode disposed in the branch passage when fluid within the branch passage is stationary.

7 . The apparatus of claim 1 , wherein the fluidics system further includes a waste passage extending from the fluidics circuit, the waste passage connected to a waste passage electrode capacitively connected to the electrode.

8 . The apparatus of claim 1 , further comprising an analyte in the reaction vessel.

9 . The apparatus of claim 8 , wherein the analyte comprise a particle having attached thereto a clonal population of nucleic acid fragments.

10 . The apparatus of claim 8 , wherein the electronic sensor is to generate the output signal in response to the analyte or an analyte reaction byproduct proximate thereto.

11 . A method of monitoring a reaction product in a reaction vessel, the method comprising:

flowing a first reagent within an apparatus, the apparatus comprising:

a reaction vessel coupled to an electronic sensor for monitoring a reaction product in the reaction vessel, wherein the reaction vessel includes a microwell array integrated with a sensor array;

a fluidics system for sequentially delivering a plurality of reagents to the reaction vessel, the fluidics system including a plurality of reagent reservoirs in fluidic communication via a plurality of flow paths with a fluidics circuit and to a common passage in fluidic communication between the fluidics circuit and the reaction vessel, a solution reservoir containing a solution in fluidic communication with the common passage via a branch passage connected with the common passage at a junction between the fluidics circuit and the reaction vessel, and a valve disposed between the solution reservoir and the junction; and

an electrode in contact with the solution within the branch passage, the electrode being disposed between the valve and the junction, the electrode being in electrical communication with the reaction vessel through fluid extending from the branch passage and through the common passage, wherein the electrode avoids direct contact with the plurality of reagents and uses the conductive solution for electrical contact, the electronic sensor generating an output signal depending on a voltage of the electrode;

flowing the solution through the branch channel and the common passage; and

flowing a second reagent within the apparatus.

12 . The method of claim 11 , wherein each flow path of the plurality of flow paths is connected to a flow path electrode capacitively connected to the electrode.

13 . The method of claim 11 , wherein the fluidics system further includes a waste passage extending from the fluidics circuit, the waste passage connected to a waste passage electrode capacitively connected to the electrode.

14 . The apparatus of claim 11 , wherein the electrode is in contact with the solution from the solution reservoir and does not contact any of the plurality of reagents.

15 . The method of claim 11 , wherein the electrode is configured to provide a reference voltage to the electronic sensor.

The method of claim 11 , wherein the reaction vessel includes a well of an array of wells disposed on an array of chemically sensitive field-effect transistor sensors, the electronic sensor comprising a chemically sensitive field-effect transistor sensor of the array of chemically sensitive field-effect transistor sensors.

17 . The method of claim 11 , wherein substantially none of the plurality of reagents contacts the electrode disposed in the branch passage when fluid within the branch passage is stationary.

18 . The method of claim 11 , wherein the fluidics system further includes a waste passage extending from the fluidics circuit, the waste passage connected to a waste passage electrode capacitively connected to the electrode.

19 . The method of claim 11 , further comprising an analyte in the reaction vessel.

20 . The method of claim 19 , wherein the electronic sensor is to generate the output signal in response to the analyte or an analyte reaction byproduct proximate thereto.
