Abstract

Disclosed herein are diketopiperazine microparticles having a specific surface area of less than about 67 m 2 /g. The diketopiperazine microparticle can be fumaryl diketopiperazine and can comprise a drug such as insulin.

Claims (13)

1. An inhalation system comprising a dry powder inhaler and dry powder formulation comprising crystalline microparticles of fumaryl diketopiperazine having a specific surface area of between about 35 m 2 /g and about 67 m 2 /g, and a prostaglandin in an amount of about 0.01% to about 20% by weight of the microparticles adsorbed to the microparticles, wherein the inhaler has a flow path passing through an area of powder containment and a flow path bypassing the area of powder containment, wherein in use 10-70% of the total flow exiting the inhaler passes through the area of powder containment and 90-30% of the total flow existing the inhaler bypasses the area of powder containment.

2. The inhalation system of claim 1 , wherein the prostaglandin is PG-I2 or an analog thereof.

3. The inhalation system of claim 1 , wherein the dry powder formulation is provided in a unit dose cartridge.

4. The inhalation system of claim 1 , wherein the dry powder formulation is provided prefilled in the inhaler.

5. The inhalation system of claim 4 , wherein the inhaler comprises a mouthpiece, wherein the flow through the powder containment area and the flow bypassing the powder containment area combine prior to exiting the mouthpiece.

6. The inhalation system of claim 1 , comprising 1 to 30 mg of said dry powder formulation.

7. The inhalation system of claim 5 , wherein greater than 75% of the dry powder formulation is dispensed when the flow rate through the inhaler is from 7 to 70 liters per minute.

8. The inhalation system of claim 6 , wherein greater than 85% of the dry powder formulation is dispensed.

9. The inhalation system of claim 1 , wherein the respirable fraction/fill is greater than 40%.

10. The inhalation system of claim 9 , wherein the respirable fraction/fill is greater than 50%.

11. The inhalation system of claim 9 , wherein the respirable fraction/fill is greater than 60%.

12. An inhalation system comprising a dry powder formulation comprising crystalline microparticles of fumaryl diketopiperazine, having a specific surface area of between about 35 m 2 /g and about 67 m 2 /g, and a prostaglandin adsorbed to the microparticles, and a dry powder inhaler comprising a mouthpiece and a powder containment area wherein the inhaler has a flow path passing through the powder containment area and a flow path bypassing the powder containment area wherein the flow through the powder containment area and the flow bypassing the powder containment area combine prior to exiting the mouthpiece.

13. The inhalation system of claim 12 , wherein the prostaglandin is PG-I2.
