Abstract

The present disclosure relates to fluidic systems and devices for processing, extracting, or purifying one or more analytes. These systems and devices can be used for processing samples and extracting nucleic acids, for example by isotachophoresis. In particular, the systems and related methods can allow for extraction of nucleic acids, including non-crosslinked nucleic acids, from samples such as tissue or cells. The systems and devices can also be used for multiplex parallel sample processing.

Claims (24)

1. A kit comprising:

(a) microfluidic chip comprising two or more fluidic channels each in fluid communication with a sample reservoir configured to receive a sample, a first buffer reservoir configured to receive a buffer, a second buffer reservoir configured to receive a second buffer, a third buffer reservoir configured to receive a third buffer and an elution reservoir configured to receive an eluate; and

(b) one or more buffers.

2. The kit according to claim 1 , wherein the one or more buffers comprises a leading electrolyte buffer.

3. The kit according to claim 1 , wherein the one or more buffers comprises a trailing electrolyte buffer.

4. The kit according to claim 1 , wherein the one or more buffers comprises a sample buffer.

5. The kit according to claim 1 , wherein the one or more buffers comprises a lysis buffer.

6. The kit according to claim 1 , wherein the one or more buffers comprises a leading electrolyte buffer, a trailing electrolyte buffer, a sample buffer and a lysis buffer.

7. The kit according to claim 6 , wherein the kit further comprises one or more enzymes.

8. The kit according to claim 7 , wherein the one or more enzymes comprises one or more of an RNase, a DNase and a proteinase.

9. The kit according to claim 1 , wherein the two or more fluidic channels each comprise a branched fluidic channel comprising a loading section and an isotachophoresis (“ITP”) section wherein the ITP section comprises a leading electrolyte branch and an elution branch, meeting at a branch point.

10. The kit according to claim 9 , wherein the loading section comprises:

(1) a trailing electrolyte well fluidically connected to a sample input region of the fluidic channel; and

(2) a sample input well fluidically connected to the sample input region of the fluidic channel.

11. The kit according to claim 10 , wherein the each fluidic channel further comprises a capillary barrier positioned between the sample input region of the fluidic channel and the ITP section.

12. The kit according to claim 10 , wherein the loading section further comprises a trailing capillary barrier positioned between the trailing electrolyte well and the sample input well, and a gas channel connected to the trailing capillary barrier and to a pneumatic pressure port.

13. The kit according to claim 9 , wherein the leading electrolyte branch comprises a leading electrolyte well fluidically connected to the fluidic channel.

14. The kit according to claim 9 , wherein the elution branch comprises a sample elution well fluidically connected to the fluidic channel.

15. The kit according to claim 9 , wherein the ITP section further comprises a leading electrolyte buffering well fluidically connected to the leading electrolyte well through a leading electrolyte buffering channel.

16. The kit according to claim 15 , wherein the leading electrolyte buffering channel comprises a leading capillary barrier and a gas channel connected to the leading capillary barrier and to a pneumatic pressure port.

17. The kit according to claim 9 , wherein the elution branch further comprises an elution buffering well fluidically connected to the elution well through an elution buffering channel.

18. The kit according to claim 17 , wherein the elution buffering channel comprises an elution capillary barrier, and a gas channel connected to the elution capillary barrier and to a pneumatic pressure port.

19. The kit according to claim 17 , wherein the elution buffering channel comprises an elution capillary barrier, and a gas channel connected to the elution capillary barrier and to a pneumatic pressure port.

20. The kit according to claim 1 , wherein the kit comprises 8 fluidic channels.
