Abstract

Disclosed herein are methods for treating a cancer comprising: a. administering a Btk inhibitor to a subject sufficient to result in an increase or appearance in the blood of a subpopulation of lymphocytes defined by immunophenotyping; b. determining the expression profile of one or more biomarkers from one or more subpopulation of lymphocytes; and c. administering a second agent based on the determined expression profile.

Claims (4)

1 . A method of inhibiting Bruton's tyrosine kinase (BTK) activity in an individual having a B-cell malignancy comprising:

orally administering to the individual a dose of an irreversible BTK inhibitor;

wherein the dose is an oral dose determined to result in at least about a 90% mean steady state occupancy of the BTK active sites in peripheral blood mononuclear cells in a population of individuals having the B-cell malignancy.

2 . The method of claim 1 , wherein the dose is a dose determined to result in about 100% mean steady state occupancy of the BTK active sites in peripheral blood mononuclear cells in a population of individuals having the B-cell malignancy.
