Physiologically-tailored tissue organoids are disclosed for monitoring and treating diseases and improving an individual's health.

29 . A method of treating an individual in need thereof for a disease, disorder, or addiction, comprising:
contacting a tissue organoid to the individual, the tissue organoid comprising a population of genetically engineered cells comprising at least one recombinant gene encoding a therapeutic agent.
30 . The method of claim 29 , wherein the tissue organoid is biointegrated into the individual by grafting or surgical implantation.
31 . The method of claim 29 , wherein the disease or disorder is PKU or hemophilia, and
wherein the addiction is one or more of cocaine addiction, alcoholism, nicotine addiction, or amphetamine addiction.

50 . The method of claim 29 , wherein the therapeutic agent is expressed constitutively or by induction with an inducer.
51 . The method of claim 29 , wherein the therapeutic agent comprises phenylalanine ammonia lyase (PAL).
52 . The method of claim 51 , wherein the therapeutic agent treats phenylketonuria (PKU) in the individual.
53 . The method of claim 29 , wherein the therapeutic agent comprises Factor VIII or Factor IX.
54 . The method of claim 53 , wherein the therapeutic agent is conjugated with albumin.
55 . The method of claim 54 , wherein the therapeutic agent treats hemophilia in the individual.
56 . The method of claim 29 , wherein the therapeutic agent comprises hBChE.
57 . The method of claim 56 , wherein the therapeutic agent treats cocaine addiction in the individual.
58 . The method of claim 29 , wherein the therapeutic agent comprises one or more of DkTx, VaTx, glucagon-like peptide 1 (GLP-1), a GLP-1 analog, a modified GLP-1 (mGLP1), and a mGLP1 analog.
59 . The method of claim 58 , wherein the therapeutic agent is inducible by alcohol.
60 . The method of claim 59 , wherein the therapeutic agent treats alcoholism in the individual.
61 . The method of claim 58 , wherein the therapeutic agent treats nicotine or amphetamine addiction in the individual.
62 . The method of claim 29 , wherein the therapeutic agent comprises PYY.
63 . The method of claim 62 , wherein the therapeutic agent treats obesity in the individual.
64 . The method of claim 62 , wherein the therapeutic agent causes decreased food seeking and consumption by the individual.
65 . A method of producing anti-aging effects in an individual in need thereof, comprising:
contacting a tissue organoid to the individual, the tissue organoid comprising a population of genetically engineered cells comprising at least one recombinant gene encoding a therapeutic agent, wherein the therapeutic agent comprises TIMP2 or an analog thereof.
66 . A method of treating an individual in need thereof for a disease, disorder, or addiction, comprising:
contacting a tissue organoid to the individual, the tissue organoid comprising a population of genetically engineered cells comprising at least one recombinant gene encoding a therapeutic agent, wherein expression of the therapeutic agent is inducible by an inducer; and
administering an inducer to the individual to induce expression of the therapeutic agent.
