Abstract

Embodiments of the present invention provide a method of treating a stricture in a nonvascular body lumen such as urethral strictures, benign prostatic hyperplasia (BPH) strictures, ureteral strictures, esophageal strictures, sinus strictures, and biliary tract strictures. Embodiments of the present invention provide a method for treating at least one of benign prostatic hyperplasia (BPH), prostate cancer, asthma, and chronic obstructive pulmonary disease (COPD). The method can include delivering, for example, via drug coated balloon catheters, anti-inflammatory and anti-proliferative drugs (e.g., rapamycin, paclitaxel, and their analogues) and one or more additives.

Claims (27)

1. A method of treating a nonvascular body lumen stricture, the method comprising:

contacting a coating on a medical device in a body lumen with walls of the body lumen stricture, the coating comprising at least one additive and an initial drug load of a therapeutic agent comprising paclitaxel, a paclitaxel analogue, docetaxel, a docetaxel analogue, taxol, a taxol analogue, rapamycin, a rapamycin analogue, everolimus, an everolimus analogue, tacrolimus, a tacrolimus analogue, or a combination thereof, wherein the coating is flushed or soaked in a solution comprising water or saline.

2. The method of claim 1 , wherein the medical device comprises a balloon catheter or a stent.

3. The method of claim 1 , wherein the coating is soaked in the solution outside the body.

4. The method of claim 1 , wherein the coating is soaked in the solution in the body lumen.

5. The method of claim 1 , wherein the coating is flushed or soaked in the solution prior to the contacting of the coating with the walls of the body lumen stricture.

6. The method of claim 1 , wherein the coating is flushed or soaked in the solution after the contacting of the coating with the walls of the body lumen stricture.

7. The method of claim 1 , further comprising prior the contacting of the coating with the walls of the body lumen stricture:

inserting an uncoated balloon catheter into the body lumen stricture;

inflating the uncoated balloon catheter until the balloon contacts the walls of the body lumen stricture;

deflating the uncoated balloon; and

withdrawing the uncoated balloon from the body lumen.

8. The method of claim 1 , wherein the flushing or soaking in the solution in combination with the contacting of the coating with the stricture is sufficient to release 37% to 97% of the initial drug load.

9. The method of claim 1 , wherein the coating is flushed or soaked in the solution for a time of at least 1 minute.

10. The method of claim 1 , wherein the body lumen stricture comprises a stricture in a urethra, a ureter, a prostatic urethra, an esophagus, a biliary tract, a stomach, a colon, an intestine, a sinus, a trachea, or an airway.

11. The method of claim 1 , wherein the body lumen stricture comprises a stricture in a prostatic urethra.

12. The method of claim 1 , wherein the body lumen stricture comprises a urethral stricture, a benign prostatic hyperplasia (BPH) stricture, or a ureteral stricture.

13. The method of claim 1 , wherein a ratio by weight of the therapeutic agent to the total weight of the additive in the coating is from 2 to 6, and wherein the initial drug load of the therapeutic agent is from 1 microgram to 20 micrograms per square millimeter of the medical device.

14. The method of claim 1 , wherein the additive in the coating comprises one or more water-soluble additives.

15. The method of claim 1 , wherein the additive in the coating comprises a surfactant.

16. The method of claim 1 , wherein the additive in the coating comprises a protein.

17. The method of claim 1 , wherein the additive in the coating comprises pentaerythritol ethoxylate, pentaerythritol propoxylate, pentaerythritol propoxylate/ethoxylate, or combinations thereof.

18. The method of claim 1 , further comprising visualizing the medical device comprising the coating in the body lumen stricture with a scope during the contacting.

19. The method of claim 1 , wherein the contacting of the coating on the medical device in the body lumen with walls of the body lumen stricture comprises dilating the body lumen stricture to a desired size.

20. A method of treating benign prostatic hyperplasia (BPH), the method comprising:

contacting a coating on a medical device in a prostatic urethra with walls of the prostatic urethra, the coating comprising at least one additive and an initial drug load of a therapeutic agent comprising paclitaxel, a paclitaxel analogue, docetaxel, a docetaxel analogue, taxol, a taxol analogue, rapamycin, a rapamycin analogue, everolimus, an everolimus analogue, tacrolimus, a tacrolimus analogue, or a combination thereof, wherein the coating is flushed or soaked in a solution comprising water or saline; and

visualizing the medical device comprising the coating in the prostatic urethra with a scope during the contacting.
