Abstract

Devices, systems, compositions and methods for long term or prolonged transdermal administration of an active agent are provided.

Claims (28)

1. A method for treating Alzheimer's disease in a subject, comprising:

applying a transdermal patch to skin of the subject, wherein the transdermal patch is comprised of

a composite backing layer comprised of

(i) a first layer comprised of a polymer fabric or a polymer film comprised of one or more polymers selected from polyesters, and polyethylenes;

(ii) a second layer comprised of one or more adhesive polymers, the one or more adhesive polymers being acrylate copolymers; and

(iii) a third layer comprised of a polyethylene terephthalate/ethylene-vinyl acetate laminate;

wherein the first and second layers are in contact and the second and third layers are in contact, and

wherein the third layer comprises cuts formed at least partially through the third layer; and

an adhesive matrix drug layer laminated to the third layer, the adhesive matrix drug layer comprising donepezil.

2. The method of claim 1 , wherein the first layer has a thickness of about 10-25 mil.

3. The method of claim 1 , wherein one or more of the adhesive polymers of the second layer has at least one of (i) a tensile strength of less than 10 MPa and (ii) an elongation of at least 50%.

4. The method of claim 1 , wherein the second layer has a thickness of about 0.5-30 mil.

5. The method of claim 1 , wherein the third layer has a thickness of about 1-40 mil.

6. The method of claim 1 , wherein the first layer, second layer, and third layer are laminated.

7. A method for delivery of donepezil through skin, comprising:

applying a transdermal patch to the skin of a subject, wherein the transdermal patch is comprised of a composite backing layer comprising

(i) a first layer comprised of a polymer fabric or a polymer film comprised of one or more polymers selected from polyesters, and polyethylenes;

(ii) a second layer comprised of one or more adhesive polymers, the one or more adhesive polymers being acrylate copolymers; and

(iii) a third layer comprised of a polyethylene terephthalate/ethylene-vinyl acetate laminate,

wherein the third layer comprises cuts formed at least partially through the third layer;

wherein the first layer, second layer, and third layers are arranged in contact as a composite;

(iv) an adhesive drug layer comprising donepezil, the adhesive drug layer laminated to the third layer; and

(v) a release liner;

wherein donepezil is delivered through the skin upon removal of the release liner.

8. The method of claim 7 , wherein the first layer has a thickness of about 10-25 mil.

9. The method of claim 7 , wherein the second layer has a thickness of about 0.5-30 mil.

10. The method of claim 7 , wherein the third layer has a thickness of about 1-40 mil.

11. The method of claim 7 , wherein the release liner is formed of a material selected from a silicone coated polyethylene terephthalate, a fluorocarbon, a fluorocarbon coated polyethylene terephthalate, and a fluorosilicone coated polyethylene terephthalate.
