Treatment of immunosuppressed subjects
An IL-15 super agonist (IL-15N72D:IL-15RαSU/IgG1Fc; N-803) increases circulating NK cells, effector memory and effector memory RA cells in post-allogeneic hematopoietic stem cell transplant patients (HCT). Methods of treatment include administration of N-803 to subjects in need of such treatment.
1. A kit for use in treating human immunodeficiency virus (HIV) latent infection in a subject in need thereof, the kit comprising:
a. isolated immune effector cells, stem cells, or a combination thereof, wherein the immune effector cells are selected from the group consisting of natural killer cells (NK) cytolytic CD8 + T cells (CTLs), CD4 + T helper cells (T H ), effector memory (T EM ) T cells (CD45RA − /CCR7 − ), effector memory RA (T EMRA ) T cells (CD45RA + /CCR7 − ), and combinations thereof;
b. a composition comprising from about 1 mcg/kg up to 6.0 mcg/kg of an IL-15:IL-15Rα complex wherein the IL-15:IL-15Rα complex is an IL-15N72D:IL-15RαSu/Fc complex comprising a dimeric IL-15RαSu/Fc and two IL-15N72D molecules;
c. an adjuvant, wherein the adjuvant is a synthetic toll-like receptor-7/8 (TLR7/8) agonist; and
d. instructions for using the kit.
2. The kit of claim 1 , wherein the kit further comprises a carrier selected from the group consisting of a box, carton, and tube.
3. The kit of claim 1 , wherein the kit further comprises a container selected from the group consisting of vials, tubes, ampoules, bottles, syringes, and bags.
4. The kit of claim 1 further comprising one or more chemotherapeutic agents, cytokine antagonist, cytokine receptor antagonist, cytokines, adoptive cell therapy, anti-viral agents, checkpoint inhibitors, or combinations thereof.

The kit of claim 4 , wherein the anti-viral agent comprises: a non-nucleoside reverse transcriptase inhibitor (NNRTI) selected from the group consisting of etravirine, efavirenz, nevirapine, rilpivirine, delavirdine, and combinations thereof; a nucleoside reverse transcriptase inhibitor (NRTI) selected from the group consisting of lamivudine, zidovudine, emtricitabine, abacavir, zalcitabine, dideoxycytidine, azidothymidine, tenofovir disoproxil fumarate, didanosine (ddI EC), dideoxyinosine, stavudine, abacavir sulfate, and combinations thereof; a protease inhibitor selected from the group consisting of amprenavir, tipranavir, indinavir, saquinavir mesylate, lopinavir and ritonavir (LPV/RTV), Fosamprenavir Calcium (FOS-APV), ritonavir, darunavir, atazanavir sulfate, nelfinavir mesylate, and combinations thereof; or combinations thereof.
6. The kit of claim 1 further comprising a pharmaceutically acceptable buffer.
7. The kit of claim 6 , wherein the buffer is physiological saline.
8. The kit of claim 1 , wherein the composition is in a dosage form suitable for parenteral administration.
9. The kit of claim 1 , wherein the composition is in the form of a solution, a suspension, an emulsion or is presented as a dry powder to be reconstituted in a parentally acceptable liquid vehicle and/or solvent.
10. The kit of claim 9 , wherein the parentally acceptable liquid vehicle and/or solvent is selected from the group consisting of water, 1,3-butanediol, Ringer's solution, isotonic sodium chloride solution and dextrose solution.
11. The kit of claim 10 , wherein the water is adjusted to a suitable pH by addition of an appropriate amount of hydrochloric acid, sodium hydroxide or a suitable buffer.
12. The kit of claim 1 , wherein the composition is formulated with an excipient, carrier and/or a preservative.
13. The kit of claim 1 , wherein the composition is in a unit dosage form.
14. The kit of claim 1 , wherein the composition is incorporated into microspheres, microcapsules, nanoparticles, or liposomes.
15. The kit of claim 1 , wherein the kit further comprises a histone deacetylase (HDAC) inhibitor.
