Abstract

The present invention is drawn to methods and compositions for treating inflammatory disorders of the lower airways, comprising administering an effective amount of an agent, which modulates the expression and/or activity of a proinflammatory cytokine or fragment thereof, preferably in a human. The proinflammatory cytokine contemplated by the invention includes IL-1, IL-6, IL-8 and TNF-alpha. The present invention describes a kit comprising a delivery device and a pharmaceutical composition for administration of the agent. The pharmaceutical composition includes at least one proinflammatory cytokine inhibitor, optionally one or more additional active ingredients, and at least one pharmaceutically active carrier. The delivery device further comprises a nebulizer, an inhaler, a powder dispenser, an intrapulmonary aerosolizer and a sub-miniature aerosolizer.

Claims (20)

1 . A method for treating an inflammatory disorder of lower airways in a human subject in need thereof, comprising administering directly to the lower airways in the human subject an effective amount of anakinra, wherein the effective amount of anakinra is 0.01 mg to 100 mg per day.

2 . The method of claim 1 , wherein the inflammatory disorder of the lower airways is chronic obstructive pulmonary disease (COPD).

3 . The method of claim 1 , wherein the inflammatory disorder of the lower airways is lung injury.

4 . The method of claim 1 , wherein anakinra is administered via inhalation or via direct instillation into the lower airways.

5 . The method of claim 1 , wherein the effective amount of anakinra is 0.1 mg to 50 mg per administration.

6 . The method of claim 1 , wherein the effective amount of anakinra is 0.5 mg to 5.0 mg per administration.

7 . The method of claim 1 , wherein the inflammatory disorder of the lower airways is cystic fibrosis.

8 . The method of claim 1 , wherein the inflammatory disorder of the lower airways is bronchiolitis obliterans syndrome.

9 . The method of claim 1 , wherein the inflammatory disorder of the lower airways is allograft rejection.

10 . The method of claim 1 , wherein the method further includes administering a pharmaceutical carrier.

11 . The method of claim 1 , wherein the pharmaceutical carrier is selected from the group consisting of saline, Ringer's solution, dextrose solution, and a combination thereof.

12 . The method of claim 1 , wherein the method comprising administering two or more dosages of anakinra in less than 5 minutes apart.

13 . The method of claim 1 , wherein the effective amount of anakinra is delivered using a device selected from the group consisting of a metered dose inhaler, a sprayer, a nebulizer, and a dry powder generator.

14 . The method of claim 1 , wherein the effective amount of anakinra is delivered using a nebulizer.

15 . The method of claim 1 , wherein the nebulizer is selected from the group consisting of an AERX Aradigm nebulizer, a Ultravent nebulizer (Mallinckrodt), and an Acorn II nebulizer.

16 . The method of claim 1 , wherein the device releases a mixture containing anakinra and having an average particle size of less than 20 microns in diameter.

17 . The method of claim 1 , wherein the device releases a mixture containing anakinra and having an average particle size of less than 10 microns in diameter.

18 . The method of claim 1 , wherein the device releases a mixture containing anakinra and having an average particle size of less than 5 microns in diameter.

19 . The method of claim 1 , wherein the device releases a mixture containing anakinra and having an average particle size of between 1 micron and 5 microns in diameter.

20 . The method of claim 1 , wherein the device releases a mixture containing anakinra and having an average particle size of between 3 microns and 5 microns in diameter.
