Riluzole prodrugs and their use
Pharmaceutical compositions of the invention include substituted riluzole prodrugs useful for the treatment of cancers including melanoma, breast cancer, brain cancer, and prostate cancer through the release of riluzole. Prodrugs of riluzole have enhanced stability to hepatic metabolism and are delivered into systemic circulation by oral administration, and then cleaved to release riluzole in the plasma via either an enzymatic or general biophysical release process.
1 . A method for treating spinocerebellar ataxia in a subject in need of such treatment, comprising administering a riluzole prodrug to the subject, wherein said administration comprises:
releasing an effective amount of riluzole or hydrate, solvate, pharmaceutically acceptable salt, or complex thereof into blood stream of the subject; and
attenuating presynaptic glutamate release in the subject, and
wherein the prodrug is a compound that is cleaved to release riluzole in the blood stream via either an enzymatic or general biophysical release process.
2 . The method according to claim 1 , wherein the spinocerebellar ataxia is hereditary spinocerebellar ataxia.
3 . A method for treating spinocerebellar ataxia in a subject in need of such treatment, comprising administering a riluzole prodrug to the subject, wherein said administration comprises:
contacting plasma of the subject with an effective amount of riluzole or hydrate, solvate, pharmaceutically acceptable salt, or complex thereof; and
attenuating presynaptic glutamate release in the subject, and
wherein the prodrug is a compound that is cleaved to release riluzole in the plasma via either an enzymatic or general biophysical release process.
4 . The method according to claim 3 , wherein the spinocerebellar ataxia is hereditary spinocerebellar ataxia.
