Abstract

The present invention relates to fibers comprising polyesteramide co-polymers and a bioactive agent. The present invention also relates to the fibers for use in medical applications especially for use in the delivery of bioactive agents. In an embodiment, a fiber degrades hydrolytically at physiological conditions via bulk erosion.

15 . A method comprising the step of treating a condition associated with the eye in a patient suffering therefrom by degrading hydrolytically via bulk erosion a fiber implanted in the patient, the fiber comprising a bioactive agent and a biodegradable poly(esteramide) copolymer (PEA) according to structural Formula (IV),

wherein

m+p is from 0.9 to 0.1 and q is from 0.1 to 0.9;

m+p+ q =1, whereby m and p are each greater than 0;

n is about 5 to about 300;

a is at least 0.05, b is at least 0.05, a+b=1, qa=q*a, and qb=q*b; wherein units of m, units of p, units of q a, and units of q b are all randomly distributed throughout the copolymer;

R 1 is (C 2 -C 20 ) alkylene;

R 3 and R 4 may be the same or different and are each selected from the group consisting of hydrogen, —CH 3 , —CH(CH 3 ) 2 , —CH(CH 3 )—CH 2 —CH 3 , —CH 2 —CH(CH 3 ) 2 , —CH 2 —C 6 H 5 , (CH 3 ) 2 CH—CH 2 —, and —(CH 2 ) 2 S(CH 3 );

R 5 is (C 2 -C 20 )alkylene;

R 6 is according to structural Formula (III);

R 7 is (C 6 -C 10 )aryl(C 1 -C 6 )alkylene; and

R 8 is (C 2 -C 20 )alkylene.

16 . The method according to claim 15 , wherein a is at least 0.15.

17 . The method according to claim 15 , wherein a is at least 0.5.

18 . The method according to claim 15 , wherein a is at least 0.75.

19 . The method according to claim 15 , wherein a is at least 0.8.

20 . The method according to claim 15 , wherein n is from 50 to 200.

21 . The method according to claim 15 , wherein q is from 0.1 to 0.25.

22 . The method according to claim 15 , wherein R 3 and R 4 are the same.

23 . The method according to claim 15 , wherein R 3 and R 4 may be the same or different and are each selected from the group consisting of hydrogen, —CH 3 , —CH(CH 3 ) 2 , —CH(CH 3 )—CH 2 —CH 3 , —CH 2 —CH(CH 3 ) 2 , —CH 2 -C 6 H 5 , and (CH 3 ) 2 CH—CH 2 —.

24 . The method according to claim 15 , wherein R 5 is —(CH 2 ) 6 —.

25 . The method according to claim 15 , wherein R 5 is —(CH 2 ) 4 —.

26 . The method according to claim 15 , wherein R 3 and R 4 are each (CH 3 ) 2 CH—CH 2 —, and R 7 is benzyl.

27 . The method according to claim 15 , wherein R 1 is —(CH 2 ) 8 —, R 3 and R 4 are each (CH 3 ) 2 CH—CH 2 —, and R 7 is benzyl.

28 . The method according to claim 15 , wherein R 1 is —(CH 2 ) 8 —, R 3 and R 4 are each (CH 3 ) 2 CH—CH 2 —, R 7 is benzyl, and R 8 is —(CH 2 ) 4 —.

29 . The method according to claim 15 , wherein qa=0.19 and qb=0.06.

30 . The method according to claim 15 , wherein m=0.3; p=0.45; qa=0.19; and qb=0.06.

31 . The method according to claim 15 , wherein the bioactive agent comprises a prostaglandin or a prodrug thereof.

The method according to claim 15 , wherein the fiber further comprises a poly(ortho ester), poly(anhydride), poly(D,L-lactic acid), poly (L-lactic acid), poly(glycolic acid), copolymers of poly(lactic) and glycolic acid, poly(L-lactide), poly(D,L-lactide), poly(glycolide), poly(D,L-lactide-co-glycolide), or poly(L-lactide-co-glycolide), poly(phospho esters), poly(trimethylene carbonate), poly(oxa-esters), poly(oxa-amides), poly(ethylene carbonate), poly(propylene carbonate), poly(phosphoesters), poly(phosphazenes), poly(tyrosine derived carbonates), poly(tyrosine derived arylates), poly(tyrosine derived iminocarbonates), or combinations thereof.

33 . The method according to claim 15 , wherein the average diameter of the fiber is from 50 to 500 micrometers.

34 . The method according to claim 15 , wherein the fiber comprises a first length and first diameter prior to implantation and a second length and second diameter after implantation, wherein the first length is greater than the second length and the first diameter is less than the second diameter.
