Abstract

Disclosed herein are mutant photosynthetic microorgnaisms having an attenuated SGI1 gene. The mutants have reduced chlorophyll and increased productivity with respect to wild type cells. Also disclosed are methods of using such mutants for producing biomass or bioproducts, and methods of screening for such mutants.

Claims (20)

1 . A mutant Chlorophyte algal organism having an attenuated gene encoding an SGI1 polypeptide having at least 90% sequence identity to a SEQ ID NO: 17; and wherein the photosynthetic organism has at least 25% higher lipid productivity than a control organism grown or cultured under the substantially the same conditions.

2 . The mutant Chlorophyte algal organism of claim 1 wherein the attenuation is a deletion or a disruption in the gene encoding the SGI1 polypeptide.

3 . The mutant Chlorophyte algal organism of claim 2 wherein the gene encoding the SGI1 polypeptide has at least 95% sequence identity to a SEQ ID NO: 17.

4 . The mutant Chlorophyte algal organism of claim 1 wherein the algal organism has at least 40% higher lipid productivity than the control organism.

5 . The mutant photosynthetic organism according to claim 1 , wherein the mutant photosynthetic organism has reduced chlorophyll with respect to the control photosynthetic microorganism.

6 . The mutant photosynthetic organism according to claim 4 , wherein the mutant photosynthetic organism has an increased chlorophyll a:b ratio with respect to the control photosynthetic organism.

7 . The mutant photosynthetic organism according to claim 1 , wherein the mutant photosynthetic organism has a reduced photosystem II (PSII) antenna size with respect to the control photosynthetic organism.

8 . The mutant photosynthetic organism according to claim 1 , wherein the mutant photosynthetic organism has a higher 14C Pmax with respect to the control photosynthetic organism.

9 . The mutant photosynthetic organism according to claim 1 , wherein the mutant photosynthetic organism has a higher Fv/Fm than the control organism.

10 . The mutant photosynthetic organism according to claim 1 , wherein the mutant photosynthetic organism has a higher protein content than the control organism.

11 . The mutant algal microorganism according to claim 1 , wherein the mutant algal microorganism is a Chlorophyte of the Class Trebouxiophyceae.

12 . The mutant algal microorganism according to claim 11 , wherein the mutant algal microorganism is a species of the genus Oocystis.

13 . The mutant algal microorganism according to claim 12 , wherein the cross-sectional unit size of the PSII antenna size is reduced by at least 25%, with respect to the cross-sectional unit size of the PSII antenna of the control microorganism.

14 . The mutant algal microorganism according to claim 12 , wherein the Pmax (14C) is increased by at least 25% with respect to the Pmax (14C) of the control microorganism.

15 . The mutant algal microorganism according to claim 1 , wherein the mutant algal microorganism has at least 20% less chlorophyll with respect to the control algal microorganism.

16 . The mutant algal microorganism according to claim 1 , wherein the mutant algal microorganism has at least 20% less chlorophyll b with respect to the control algal microorganism.

17 . The mutant algal microorganism according to claim 16 , wherein the mutant algal microorganism has at least 20% more protein as a percent of total organic carbon than the control algal microorganism.

18 . The mutant algal microorganism according to claim 1 , wherein the mutant algal microorganism has reduced expression of at least 10 LHC genes with respect to the control algal microorganism.

19 . A method of making an lipid product comprising culturing the algal mutant of claim 1 to produce the lipid product.

20 . The method according to claim 19 , further comprising recovering the lipid product from the culture.
