END-TO-END CELL THERAPY AUTOMATION
The present disclosure provides an automated method of producing genetically modified immune cells, including chimeric antigen receptor T (CAR T) cells, utilizing a fully-enclosed cell engineering system.
1 . An automated cell engineering system comprising:
a) an outer shell comprising a plurality of separate chambers configured to activate, expand, concentrate and harvest a cell culture,
wherein one of the chambers comprises an electroporation unit, and one of the chambers comprises a magnetic separation unit;
b) a self-contained, removable and replaceable cassette that includes a substantially non-yielding cell culture chamber;
c) at least two of a temperature sensor, a pH sensor, a glucose sensor, an oxygen sensor, a carbon dioxide sensor, and an optical density sensor configured to monitor a temperature, a pH, a glucose concentration, and oxygen level, a carbon dioxide level, and/or an optical density of a cell culture within the self-contained, removable and replaceable cassette,
wherein the automated cell engineering system is configured to be prefilled with an activation reagent, a vector, a cell culture media, nutrients, and a selection reagent, and wherein a first chamber of the separate chambers is maintained at a temperature of about 25° C. to about 37° C., and a second chamber is maintained at a temperature of about 4° C. to about 8° C.

The system of claim 1 , further comprising a pump configured to drive fluid to and/or from the substantially non-yielding cell culture chamber in response to monitoring one or more of the temperature sensor, the pH sensor, the glucose sensor, the oxygen sensor, the carbon dioxide sensor, and the optical density sensor.
3 . The system of claim 1 , further comprising a user interface configured to receive user inputs.
4 . The system of claim 1 , further comprising a computer control system.
5 . The system of claim 1 , wherein the self-contained, removable and replaceable cassette comprises one or more sampling and/or injection ports.
6 . The system of claim 1 , wherein the substantially non-yielding cell culture chamber supports at least 2 billion cells.
