Diketopiperazine Salts for Drug Delivery and Related Methods
Drug delivery systems have been developed based on the formation of diketopiperazine carboxylate salts and microparticles containing the same. The systems may further comprise a bioactive agent. Related methods for making and using the biologically active agent delivery compositions are also provided. In certain embodiments, the pharmaceutically acceptable salts described can be formed by removal of solvent by methods including distillation, evaporation, spray drying or lyophilization.
1 . A dry powder composition comprising:
disodium fumaryl diketopiperazine (FDKP) particles chemically associated with insulin to form a disodium FDKP-insulin microparticle complex;
wherein the disodium FDKP-insulin microparticle complex is defined as having an average respirable fraction of 88% with a Standard Deviation of 1.60, a size range of between about 0.5 μm to about 10 μm, and a rugosity of less than 2.
2 . The dry powder composition of claim 1 wherein the rugosity is less than 1.
3 . The dry powder composition of claim 1 wherein the coefficient of variation is about 1.8.
4 . The dry powder composition of claim 1 wherein the disodium FDKP-insulin microparticle complex is in the size range of about 0.5 μm to about 6 μm.
5 . The dry powder composition of claim 1 wherein the disodium FDKP-insulin microparticles are about 25% insulin by weight.
6 . The dry powder composition of claim 1 wherein the disodium FDKP-insulin microparticles having a fine particle fraction of less than 5.8 μm is 49.21%.
