Abstract

Provided are compositions for treating CD96 mediated diseases, and related methods.

Claims (20)

1 . A method for the treatment of a cancer in a human in need thereof, the method comprising administering to the human a CD96 binding protein, a PD-1 antagonist, and a TIGIT antagonist, wherein the CD96 binding protein comprises CDRH1 of SEQ ID NO: 115; CDRH2 of SEQ ID NO: 145; CDRH3 of SEQ ID NO: 147; CDRL1 of SEQ ID NO: 97; CDRL2 of SEQ ID NO: 99; and CDRL3 of SEQ ID NO: 101.

2 . The method of claim 1 , wherein the CD96 binding protein comprises a VH region of SEQ ID NO: 86 and a VL region of SEQ ID NO: 85.

3 . The method of claim 1 , wherein the CD96 binding protein is an antibody.

4 . The method of claim 3 , wherein the CD96 antibody further comprises a human wild-type IgG1 Fc domain.

5 . The method of claim 4 , wherein the CD96 antibody comprises a heavy chain of SEQ ID NO: 165 and a light chain of SEQ ID NO: 166.

6 . The method of claim 1 , wherein the TIGIT antagonist is an anti-TIGIT antibody.

7 . The method of claim 6 , wherein the anti-TIGIT antibody comprises a VH region of SEQ ID NO: 362 and a VL region of SEQ ID NO: 363.

8 . The method of claim 7 , wherein the anti-TIGIT antibody is EOS884488.

9 . The method of claim 1 , wherein the PD-1 antagonist is an anti-PD-1 antibody.

10 . The method of claim 9 , wherein the anti-PD-1 antibody comprises a heavy chain sequence of SEQ ID NO: 160 and a light chain sequence of SEQ ID NO: 161.

11 . The method of claim 10 , wherein the anti-PD-1 antibody is dostarlimab.

12 . The method of claim 1 , wherein the PD-1 antagonist and TIGIT antagonist are administered to the human in a ratio of about 1.0:0.80 to about 1:0:1.2 (PD-1 antagonist:TIGIT antagonist).

13 . The method of claim 1 , wherein the CD96 binding protein, PD-1 antagonist, and the TIGIT antagonist are administered to the human simultaneously or sequentially in any order.

14 . The method of claim 1 , wherein the human is administered a pharmaceutical composition comprising the CD96 binding protein, the PD-1 antagonist, and the TIGIT antagonist.

15 . A combination comprising a CD96 binding protein, a PD-1 antagonist, and a TIGIT antagonist for use in the treatment of a cancer, wherein the CD96 binding protein comprises CDRH1 of SEQ ID NO: 115; CDRH2 of SEQ ID NO: 145; CDRH3 of SEQ ID NO: 147; CDRL1 of SEQ ID NO: 97; CDRL2 of SEQ ID NO: 99; and CDRL3 of SEQ ID NO: 101.

17 . The method of claim 1 , wherein the cancer is a solid tumor.

18 . The method of claim 1 , wherein the cancer is selected from the group consisting of ovarian cancer, lung cancer, gastric cancer, bladder cancer, colorectal cancer, liver cancer, renal cancer, and head and neck squamous cell carcinoma.

19 . The method of claim 1 , wherein the cancer is lung cancer.

20 . The method of claim 19 , wherein the lung cancer is non-small cell lung cancer (NSCLC).
