ANTIBODY-RESIN COUPLING APPARATUS AND METHODS
An antibody-resin coupling apparatus quickly and efficiently activates resin beads and couples them to antibodies, while preventing breakdown and crosslinking of the beads, thereby improving downstream column purification processes, extending the usable life of the resin beads, and increasing molecule capture efficiency of the resultant resin-antibody complexes, to allow improved isolation and purification of factor VIII molecules or other drug compounds.
1 . A resin washing apparatus comprising:
a vessel comprising a cylinder divided into an upper portion and a lower portion by a screen stretched across the cylinder, the upper portion comprising an inlet and the lower portion comprising an outlet; and
an agitator disposed within the upper portion, the agitator comprising a rotor and a rotating impeller comprising a hub, a first blade, and a second blade, the first and second blades extending in opposite directions perpendicularly from the hub, each blade having an axis transverse to the axis of the rotor upon which the hub is configured to rotate;
wherein the screen comprises a mesh material; and
wherein the agitator is configured to impart a force to a fluid in the vessel, wherein the force is directed away from the screen.
2 . The resin washing apparatus of claim 1 , wherein each blade has a fixed pitch.
3 . The resin washing apparatus of claim 1 , wherein each blade has rounded edges.
4 . The resin washing apparatus of claim 1 , wherein the agitator rotates at a speed sufficient to rapidly mix resin beads without breaking the resin beads.
5 . The resin washing apparatus of claim 1 , wherein the agitator rotates at between 10 and 50 RPM.
6 . The resin washing apparatus of claim 5 , wherein the agitator rotates at 20 RPM.
7 . The resin washing apparatus of claim 5 , wherein the agitator rotates at 35 RPM.
8 . The resin washing apparatus of claim 5 , wherein the mesh material has a pore size between about 5 μm and about 80 μm.
9 . The resin washing apparatus of claim 1 , further comprising a support beam configured to support the screen and prevent the screen from bowing.
10 . The resin washing apparatus of claim 1 , wherein the screen is removable from the vessel.
