Abstract

Fusion proteins containing a targeting sequence, exosporium protein, or exosporium protein fragment that targets the fusion protein to the exosporium of a recombinant Bacillus cereus family member are provided. The fusion proteins further comprise an enzyme having ACC deaminase activity, a phospholipase, a lipase, a xylanase, a xylosidase, a lactonase, a chitosanase, a protease, a glucanase, a phytase, an acid phosphatase, a pectinase, a mannanase, and/or an expansin protein. Also provided are recombinant Bacillus cereus family members that express the fusion proteins, exosporium fragments derived from the recombinant Bacillus cereus family members, and formulations containing the recombinant Bacillus cereus family members or exosporium fragments. Plant seeds treated with the recombinant Bacillus cereus family members, exosporium fragments, or formulations are also provided. The invention further relates to methods for stimulating plant growth and/or promoting plant health using the recombinant Bacillus cereus family members, exosporium fragments, or formulations.

8 . A fusion protein comprising:

(A) a targeting sequence, exosporium protein, or exosporium protein fragment that targets the fusion protein to the exosporium of a recombinant Bacillus cereus family member; and

(B) a glucanase, the glucanase:

comprising a xyloglucanase.

10 . A fusion protein of claim 8 , wherein the xyloglucanase comprises a xyloglucan-specific endo-beta-1,4-glucanase, a xyloglucan-specific exo-beta-1,4-glucanase, or a combination thereof.

11 . A fusion protein of claim 8 , wherein the xyloglucanase comprises a Bacillus xyloglucanase or a Paenibacillus xyloglucanase.

12 . A fusion protein of claim 10 , wherein the Bacillus xyloglucanase comprises a Bacillus licheniformis xyloglucanase or wherein the Paenibacillus xyloglucanase comprises a Paenibacillus pabuli xyloglucanase.

13 . A fusion protein of claim 8 , wherein the glucanase comprises an amino acid sequence having at least 70%, at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 98%, at least 99%, or 100% identity to SEQ ID NO: 300 or 299.

16 . A recombinant Bacillus cereus family member that expresses a fusion protein of claim 1 .

17 . Exosporium fragments derived from a recombinant Bacillus cereus family member of claim 16 .

18 . A formulation comprising a recombinant Bacillus cereus family member of claim 16 and an agriculturally acceptable carrier.

19 . A formulation comprising exosporium fragments of claim 17 and an agriculturally acceptable carrier.

20 . A plant seed treated with a recombinant Bacillus cereus family member of claim 16 .

21 . A plant seed treated with exosporium fragments of claim 17 .

22 . A method for stimulating plant growth and/or promoting plant health, comprising applying a recombinant Bacillus cereus family member of claim 16 to a plant growth medium, a plant, a plant seed, or an area surrounding a plant or a plant seed.

23 . A method for stimulating plant growth and/or promoting plant health, comprising applying exosporium fragments of claim 17 to a plant growth medium, a plant, a plant seed, or an area surrounding a plant or a plant seed.

24 . A method for delivering an enzyme to an animal, comprising feeding to the animal a recombinant Bacillus cereus family member of claim 16 , exosporium fragments derived from a recombinant Bacillus cereus family member of claim 16 , or a combination thereof.

27 . A fusion protein of claim 28 , wherein:

the fusion protein comprises a glucanase comprising an amino acid sequence having at least 70%, at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 98%, at least 99%, or 100% identity to SEQ ID NO: 300, and wherein the glucanase further comprises a signal peptide comprising SEQ ID NO: 340 at its amino terminus; or

the fusion protein comprises a glucanase comprising an amino acid sequence having at least 70%, at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 98%, at least 99%, or 100% identity to SEQ ID NO: 299, and wherein the glucanase further comprises a signal peptide comprising SEQ ID NO: 339 at its amino terminus.

28 . A fusion protein of claim 8 , wherein the glucanase further comprises a signal peptide.
