Abstract

The present invention relates to compositions comprising inhibitors of human histone methyltransferase EZH2 and one or more other therapeutic agents, particularly anticancer agents such as prednisone, and methods of combination therapy for administering to subjects in need thereof for the treatment of cancer.

21 . A method of treating follicular lymphoma, comprising administering to a human subject in need thereof Compound 44:

or a pharmaceutically acceptable salt thereof;

rituximab; and

lenalidomide.

22 . The method of claim 21 , wherein the follicular lymphoma is resistant or refractory.

23 . The method of claim 22 , wherein the subject has received at least one prior therapy.

24 . The method of claim 21 , wherein Compound 44 is administered as a mono-HBr salt.

25 . The method of claim 21 , wherein Compound 44, rituximab, and lenalidomide are each administered as separate compositions.

26 . The method of claim 21 , wherein the subject expresses a mutant EZH2.

27 . The method of claim 26 , wherein the subject expresses a Y641 mutant of EZH2.

28 . The method of claim 27 , wherein the Y641 mutant is selected from Y641F, Y641H, Y641N, and Y641S.

29 . The method of claim 26 , wherein the subject expresses a mutant EZH2 with a mutation selected from the group consisting of a substitution of glycine (G) for the wild type residue alanine (A) at amino acid position 677 of SEQ ID NO: 1 (A677G), a substitution of valine (V) for the wild type residue alanine (A) at amino acid position 687 of SEQ ID NO: 1 (A687V), a substitution of methionine (M) for or a pharmaceutically acceptable salt thereof; and

rituximab.

31 . The method of claim 30 , wherein the follicular lymphoma is resistant or refractory.

32 . The method of claim 31 , wherein the subject has received at least one prior therapy.

33 . The method of claim 31 , wherein the method further comprises administering an anticancer agent in addition to rituximab.

34 . The method of claim 30 , wherein Compound 44 is administered as a mono-HBr salt.

35 . The method of claim 30 , wherein Compound 44 and rituximab are each administered as separate compositions.

36 . The method of claim 30 , wherein the subject expresses a mutant EZH2.

37 . The method of claim 30 , wherein the subject expresses a Y641 mutant of EZH2.

38 . The method of claim 37 , wherein the Y641 mutant is selected from Y641F, Y641H, Y641N, and Y641S.

The method of claim 36 , wherein the subject expresses a mutant EZH2 with a mutation selected from the group consisting of a substitution of glycine (G) for the wild type residue alanine (A) at amino acid position 677 of SEQ ID NO: 1 (A677G), a substitution of valine (V) for the wild type residue alanine (A) at amino acid position 687 of SEQ ID NO: 1 (A687V), a substitution of methionine (M) for lenalidomide.

41 . The method of claim 40 , wherein the follicular lymphoma is resistant or refractory.

42 . The method of claim 41 , wherein the subject has received at least one prior therapy.

43 . The method of claim 41 , wherein the method further comprises administering another anticancer agent in addition to lenalidomide.

44 . The method of claim 40 , wherein Compound 44 is administered as a mono-HBr salt.

45 . The method of claim 40 , wherein Compound 44 and lenalidomide are each administered as separate compositions.

46 . The method of claim 40 , wherein the subject expresses a mutant EZH2.

47 . The method of claim 46 , wherein the subject expresses a Y641 mutant of EZH2.

48 . The method of claim 47 , wherein the Y641 mutant is selected from Y641F, Y641H, Y641N, and Y641S.

The method of claim 46 , wherein the subject expresses a mutant EZH2 with a mutation selected from the group consisting of a substitution of glycine (G) for the wild type residue alanine (A) at amino
