Abstract

The present invention provides compositions and methods for treating a disease treatable by asparagine depletion in a human subject comprising administering to a human subject a recombinant L-asparaginase.

Claims (64)

1 . A pharmaceutical composition comprising:

(i) a recombinant L-asparaginase, and

(ii) trehalose,

wherein the recombinant L-asparaginase is a tetramer,

wherein the tetramer comprises four monomers, and

wherein each monomer comprises an amino acid sequence having at least 85% sequence identity to SEQ ID NO: 1.

2 . The composition of claim 1 , wherein each monomer comprises an amino acid sequence of SEQ ID NO: 1.

3 . The composition of claim 1 , wherein the composition has a pH of about 7.0.

4 . The composition of claim 1 , wherein the recombinant L-asparaginase is conjugated to a PEG moiety.

5 . The composition of claim 1 , wherein the recombinant L-asparaginase demonstrates less than 6% aggregation.

6 . The composition of claim 1 , wherein the recombinant L-asparaginase demonstrates less than 4% aggregation.

7 . The composition of claim 1 , wherein the recombinant L-asparaginase demonstrates less than 1% aggregation.

8 . The composition of claim 1 , wherein the recombinant L-asparaginase is recombinantly produced in Pseudomonas fluorescens.

9 . The composition of claim 1 , wherein the composition comprises about 6.4% of trehalose.

10 . The composition of claim 1 , further comprising sodium phosphate.

11 . A method of treating a disease characterized by benefiting from asparagine depletion in a human subject comprising: administering to the human subject a dose of the pharmaceutical composition of claim 1 , thereby treating the disease.

12 . The method of claim 11 , wherein the recombinant L-asparaginase is administered every 48 hours in an amount of 25 mg/m 2 for two weeks.

13 . The method of claim 11 , wherein the recombinant L-asparaginase is administered every other day over a period of 5 consecutive days followed by a rest period of 2 consecutive days.

14 . The method of claim 11 , wherein the dose regimen of the pharmaceutical composition comprises a cycle.

15 . The method of claim 14 , wherein the cycle comprises a first dose, a second dose and a third dose, wherein the cycle is optionally repeatable.

16 . The method of claim 15 , wherein the first dose, the second dose, and the third dose each independently comprise about 25 mg/m 2 or about 50 mg/m 2 of the recombinant L-asparaginase.

17 . The method of claim 15 , wherein the first dose, second dose, and third dose are administered about 48-72 hours apart.

18 . The method of claim 11 , wherein the pharmaceutical composition is administered three times a week.

19 . The method of claim 11 , wherein the pharmaceutical composition is administered for 2 weeks.

20 . The method of claim 14 , wherein the first dose and the second dose are each administered in an amount of about 25 mg/m 2 .

21 . The method of claim 20 , wherein the third dose is administered in an amount of about 50 mg/m 2 .

22 . The method of claim 14 , wherein the first dose is administered on Monday, the second dose is administered on a Wednesday, and the third dose is administered on a Friday of the same week.

23 . The method of claim 14 , wherein the third dose is larger than the first dose and the second dose.

24 . The method of claim 14 , wherein the cycle is repeated at least once, the first dose and the second dose are administered about 48 hours apart, the second and the third dose are administered about 48 hours apart, and the third dose is administered about 72 hours before the first dose of a subsequent cycle.

25 . The method of claim 11 , wherein the disease is acute lymphoblastic leukemia (ALL), lymphoblastic lymphoma (LBL), colon carcinoma, colon adenocarcinoma, or colorectal adenocarcinoma.

26 . The method of claim 11 , wherein administering comprises intramuscular or intravenous administration.

27 . A method of treating cancer in a human subject comprising: administering to the human subject a dose of the pharmaceutical composition of claim 1 ,

wherein the cancer is selected from the group consisting of acute lymphoblastic leukemia (ALL), lymphoblastic lymphoma (LBL), colon carcinoma, colon adenocarcinoma, and colorectal adenocarcinoma, thereby treating cancer.

28 . The method of claim 27 , wherein the recombinant L-asparaginase is administered every 48 hours in an amount of 25 mg/m 2 for two weeks.

29 . The method of claim 27 , wherein the recombinant L-asparaginase is administered every other day over a period of 5 consecutive days followed by a rest period of 2 consecutive days.

30 . The method of claim 27 , wherein the dose regimen of the pharmaceutical composition comprises a cycle.

31 . The method of claim 30 , wherein the cycle comprises a first dose, a second dose and a third dose, wherein the cycle is optionally repeatable.

32 . The method of claim 31 , wherein the first dose, the second dose, and the third dose each independently comprise about 25 mg/m 2 or about 50 mg/m 2 of the recombinant L-asparaginase.

33 . The method of claim 31 , wherein the first dose, second dose, and third dose are administered about 48-72 hours apart.

34 . The method of claim 27 , wherein the pharmaceutical composition is administered three times a week.

35 . The method of claim 27 , wherein the pharmaceutical composition is administered for 2 weeks.

36 . The method of claim 30 , wherein the first dose and the second dose are each administered in an amount of about 25 mg/m 2 .

37 . The method of claim 36 , wherein the third dose is administered in an amount of about 50 mg/m 2 .

38 . The method of claim 30 , wherein the first dose is administered on Monday, the second dose is administered on a Wednesday, and the third dose is administered on a Friday of the same week.

39 . The method of claim 30 , wherein the third dose is larger than the first dose and the second dose.

40 . The method of claim 30 , wherein the cycle is repeated at least once, the first dose and the second dose are administered about 48 hours apart, the second and the third dose are administered about 48 hours apart, and the third dose is administered about 72 hours before the first dose of a subsequent cycle.

41 . The method of claim 27 , wherein administering comprises intramuscular or intravenous administration.

42 . A method of treating a disease characterized by benefiting from asparagine depletion in a human subject comprising: administering to the human subject a dose of the pharmaceutical composition of claim 1 ,

wherein the dose regimen of the pharmaceutical composition comprises a cycle, wherein the dose comprises about 25 mg/m2 or 50 mg/m 2 of the recombinant L-asparaginase.

43 . The method of claim 42 , wherein the recombinant L-asparaginase is administered every 48 hours in an amount of 25 mg/m 2 for two weeks.

44 . The method of claim 42 , wherein the recombinant L-asparaginase is administered every other day over a period of 5 consecutive days followed by a rest period of 2 consecutive days.

45 . The method of claim 42 , wherein the dose regimen of the pharmaceutical composition comprises a cycle.

46 . The method of claim 45 , wherein the cycle comprises a first dose, a second dose and a third dose, wherein the cycle is optionally repeatable.

47 . The method of claim 46 , wherein the first dose, the second dose, and the third dose each independently comprise about 25 mg/m 2 or about 50 mg/m 2 of the recombinant L-asparaginase.

48 . The method of claim 45 , wherein the first dose, second dose, and third dose are administered about 48-72 hours apart.

49 . The method of claim 42 , wherein the pharmaceutical composition is administered three times a week.

50 . The method of claim 42 , wherein the pharmaceutical composition is administered for 2 weeks.

51 . The method of claim 46 , wherein the first dose and the second dose are each administered in an amount of about 25 mg/m 2 .

52 . The method of claim 51 , wherein the third dose is administered in an amount of about 50 mg/m 2 .

53 . The method of claim 46 , wherein the first dose is administered on Monday, the second dose is administered on a Wednesday, and the third dose is administered on a Friday of the same week.

54 . The method of claim 46 , wherein the third dose is larger than the first dose and the second dose.

55 . The method of claim 46 , wherein the cycle is repeated at least once, the first dose and the second dose are administered about 48 hours apart, the second and the third dose are administered about 48 hours apart, and the third dose is administered about 72 hours before the first dose of a subsequent cycle.

56 . The method of claim 42 , wherein administering comprises intramuscular or intravenous administration.

57 . The method of claim 42 , wherein the disease is acute lymphoblastic leukemia (ALL), lymphoblastic lymphoma (LBL), colon carcinoma, colon adenocarcinoma, or colorectal adenocarcinoma.
