Abstract

A breath-powered, dry powder inhaler, a cartridge, and a pulmonary drug delivery system are provided. The dry powder inhaler can be provided with or without a unit dose cartridge for using with the inhaler. The inhaler and/or cartridge can be provided with a drug delivery formulation comprising, for example, a diketopiperazine and an active ingredient, including, peptides and proteins such as insulin, oxyntomodulin and glucagon-like peptide.

Claims (29)

1 . A single use dry powder inhaler comprising:

a container including a top surface and side wings, and

a mouthpiece including tracks, an inlet, an outlet and a dispensing port,

wherein the side wings are configured to interface with the tracks to allow movement of the container from a containment position to a dosing position, and

wherein the mouthpiece further includes an undersurface configured with a first surface flush with the top surface when in the containment position, and a second surface concave or hollow including the dispensing port above the top surface when in the dosing position.

2 . The single use dry powder inhaler of claim 1 , wherein movement of the container is configured to be operated manually.

3 . The single use dry powder inhaler of claim 1 , wherein the undersurface includes a bridge.

4 . The single use dry powder inhaler of claim 3 , wherein the bridge is configured to be in contact with the container for the movement from the containment position to the dosing position.

5 . The single use dry powder inhaler of claim 4 , wherein the movement is translational.

6 . The single use dry powder inhaler of claim 1 , wherein the single use dry powder inhaler is disposable.

7 . The single use dry powder inhaler of claim 1 , wherein the container is configured to hold a dry powder formulation.

8 . The single use dry powder inhaler of claim 7 , wherein the single use inhaler comprises a pre-metered, unit dose comprising the dry powder formulation within the container.

9 . The single use dry powder inhaler of claim 8 , wherein the single use inhaler is configured to deliver a single dose of the dry powder formulation.

10 . The single use dry powder inhaler of claim 9 , wherein the single dose of the dry powder formulation comprises an active agent.

11 . The single use dry powder inhaler of claim 9 , wherein the single dose of the dry powder formulation comprises a pharmaceutical.

12 . The single use dry powder inhaler of claim 9 , wherein the single dose is delivered by inhalation.

13 . The single use dry powder inhaler of claim 9 , wherein the single dose comprises one or more active agents.

14 . The single use dry powder inhaler of claim 1 , wherein the mouthpiece has an elongated shape.

15 . The single use dry powder inhaler of claim 1 , wherein the single use dry powder inhaler is breath powered.

16 . The single use dry powder inhaler of claim 1 , wherein the single use dry powder inhaler has a total resistance to flow in the dosing position ranging from 0.065 to about 0.200 (√kPa)/liter per minute.

17 . The single use dry powder inhaler of claim 1 , wherein the total resistance to flow is relatively constant across a pressure differential range between 0.5 kPa and 7 kPa.

18 . The single use dry powder inhaler of claim 1 , wherein the mouthpiece further has an internal volume extending from the inlet to the outlet greater than 0.2 cubic centimeters.

19 . An inhalation system for delivering a dry powder medicament to a pulmonary tract, comprising:

a container including a top surface, side wings, and a dry powder formulation, and

a mouthpiece including tracks, a first inlet, a second inlet, an outlet, an undersurface, and a dispensing port,

wherein the side wings are configured to interface with the tracks to allow movement of the container from a containment position to a dosing position,

wherein the undersurface is configured with a first surface flush with the top surface when in the containment position and a second surface concave or hollow including the dispensing port above the top surface when in the dosing position, and

wherein the first inlet is in communication with a first air flow pathway and the second inlet is in communication with a second air flow pathway.

20 . The inhalation system of claim 19 , wherein the first air flow pathway and the second air flow pathway meet in a substantially perpendicular manner within the mouthpiece.
