Abstract

Disclosed herein, inter alia, are multivalent nucleotide constructs and methods for making and using such constructs.

Claims (45)

1 . A compound having the formula:

wherein

R 1 is a nucleotide moiety;

R 2 is a nucleotide moiety or a fluorescent moiety;

R 3 is a bioconjugate reactive moiety;

W 1 is —O—, —NR 1A —, or —S—;

W 2 is —O—, —NR 2 A-, or —S—;

W 3 is —O—, —NR 3 A-, or —S—;

R 1A , R 2A , and R 3A are independently hydrogen or substituted or unsubstituted alkyl;

L 1 , L 2 , and L 3 and are independently covalent linkers; and

z3 is 0 or 1.

2 . The compound of claim 1 , wherein L 1 is L 101 -L 102 -L 103 ; wherein

L 101 , L 102 , and L 103 are independently a bond, —C(O)O—, —NHC(O)—, —C(O)NH—, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, substituted or unsubstituted cycloalkylene, substituted or unsubstituted heterocycloalkylene, substituted or unsubstituted arylene, or substituted or unsubstituted heteroarylene.

3 . The compound of claim 1 , wherein L 1 is

wherein n1 is an integer from 1 to 10.

4 . The compound of claim 1 , wherein L 2 is L 201 -L 202 -L 203 ; wherein

L 201 , L 202 , and L 203 are independently a bond, —C(O)O—, —NHC(O)—, —C(O)NH—, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, substituted or unsubstituted cycloalkylene, substituted or unsubstituted heterocycloalkylene, substituted or unsubstituted arylene, or substituted or unsubstituted heteroarylene.

5 . The compound of claim 1 , wherein L 2 is

wherein n2 is an integer from 1 to 10.

6 . The compound of claim 1 , wherein W 1 , W 2 , and W 3 are independently —NH—.

7 . The compound of claim 1 , wherein —W 1 -L 1 and —W 2 -L 2 are independently

The compound of claim 1 , wherein L 3 is L 301 -L 302 -L 303 ; wherein L 301 , L 302 , and L 303 are independently a bond, —C(O)O—, —NHC(O)—, —C(O)NH—, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, substituted or unsubstituted cycloalkylene, substituted or unsubstituted heterocycloalkylene, substituted or unsubstituted arylene, or substituted or unsubstituted heteroarylene.

9 . The compound of claim 1 , wherein R 3 is —N 3 , avidin, streptavidin, —NH 2 , —CN, —COOH,

10 . The compound of claim 1 , wherein z3 is 0 and R 3 is —N 3 , avidin, streptavidin, —NH 2 , —CN,

11 . The compound of claim 1 , wherein W 3 is —NH—, and L 3 -R 3 is

12 . The compound of claim 1 , wherein R 1 and R 2 independently have the formula:

wherein R 4 is hydrogen or a reversible terminator moiety.

13 . The compound of claim 1 , wherein R 1 and R 2 independently have the formula:

n1 is an integer from 1 to 10; and R 4 is hydrogen or a reversible terminator moiety.

14 . The compound of claim 1 , having the formula:

wherein n1 and n2 are independently an integer from 1 to 10.

17 . A biomolecule attached to a nucleotide construct, wherein said nucleotide construct has the formula:

wherein

R 1 is a nucleotide moiety;

R 2 is a nucleotide moiety or a fluorescent moiety;

W 1 is O, NR 1A , or

W 2 is O, NR 2A , or S;

R 1A and R 2A are independently hydrogen or substituted or unsubstituted alkyl; and

L 1 and L 2 and are independently covalent linkers.

18 . The biomolecule of claim 17 , wherein the biomolecule is avidin or streptavidin.

19 . The biomolecule of claim 17 , wherein the biomolecule is further attached to a fluorescent moiety.

20 . A method of imaging a target biomolecule, said method comprising directing an excitation beam onto a biomolecule comprising a detectable moiety and detecting a light emission from said detectable moiety, wherein said biomolecule is the biomolecule of claim 17 .
