Abstract

The present invention provides novel microfluidic substrates and methods that are useful for performing biological, chemical and diagnostic assays. The substrates can include a plurality of electrically addressable, channel bearing fluidic modules integrally arranged such that a continuous channel is provided for flow of immiscible fluids.

Claims (24)

21 . A method comprising:

providing a plurality of aqueous droplets in a microfluidic device, wherein at least one droplet of the aqueous droplets includes at least one cell;

introducing into the aqueous droplets reagents for an assay; and

imaging the at least one droplet while the aqueous droplets are spatially distributed in a two-dimensional sheet to detect a result of the assay, thereby determining a property of the at least one cell.

22 . The method of claim 21 , wherein the introducing step includes merging the at least one droplet with an assay droplet comprising an assay reagent.

23 . The method of claim 22 , wherein the merging step comprises the application of an electrical field.

24 . The method of claim 21 , wherein the assay comprises monitoring the at least one droplet to monitor a biological process in the at least one cell.

25 . The method of claim 21 , wherein the imaging step comprises detecting fluorescence from the at least one droplet.

26 . The method of claim 21 , wherein the method further comprises sorting the at least one droplet according to whether the at least one cell contains or produces a target analyte.

27 . The method of claim 21 , wherein the aqueous droplets contain a plurality of cells, and the method includes sorting the cells according to whether they contain or produce a particular target analyte.

28 . The method of claim 27 , wherein the assay comprises labeling the target analyte with a reporter that produces a detectable signal.

29 . The method of claim 28 , wherein the reporter comprises a chemically or fluorescently labeled antibody.

30 . The method of claim 21 , wherein the assay comprises an immunoassay.

31 . The method of claim 21 , wherein the method further comprises sorting the at least one droplet by a phenotype of the at least one cell.

32 . The method of claim 21 , wherein the aqueous droplets contain a plurality of cells, and the method includes sorting the cells by phenotype.

33 . The method of claim 21 , wherein the droplets are spatially distributed in the two-dimensional sheet within a microscopic field-of-view.

34 . The method of claim 21 , wherein the imaging comprises fluorescence imaging.

35 . The method of claim 21 , further comprising, prior to the introducing step, forming the plurality of the droplets using at least one channel of the microfluidic device.

36 . The method of claim 21 , wherein the microfluidic device comprises electrodes operable to manipulate the droplets.

37 . The method of claim 21 , wherein the microfluidic device comprises a control system operable with a sorting module to divert one droplet of the plurality of aqueous droplets to a waste module.

38 . The method of claim 21 , wherein the imaging comprises imaging fluorescence produced by one or more fluorescently labeled antibodies.

39 . The method of claim 21 , further comprising applying a force to at least one droplet of the plurality of droplets, wherein the force is selected from the group consisting of electrophoresis, dielectrophoresis, optical tweezers, cell trapping, and any combination thereof.

40 . The method of claim 21 , further comprising mechanically, electrically or optically controlling flow of at least one droplet of the plurality of droplets.
