Abstract

The present disclosure provides methods to improve the viability of an organ, or organs, by continuously administering a composition comprising NO x gas directly to the organ(s).

Claims (39)

1 . A method to improve viability of an organ, the method comprising:

administering an initial dose of a nitric oxide gas to the organ;

administering an increased dose of the nitric oxide gas to the organ;

monitoring a methemoglobin level of the organ; and

administering a reduced dose of the nitric oxide gas when the methemoglobin level meets or exceeds a threshold methemoglobin level,

wherein the initial dose, the increased dose, and the reduced dose do not exceed a concentration of 50 ppm of the nitric oxide gas, and

wherein the initial dose, the increased dose, and the reduced dose are administered for a combined time of twelve hours or less.

2 . The method of claim 1 , wherein the threshold methemoglobin level is about 5% to about 10%.

3 . The method of claim 1 , wherein the threshold methemoglobin level is about 10% to about 15%.

4 . The method of claim 1 , further comprising monitoring NO 2 and/or NO 3 levels.

5 . The method of claim 1 , wherein the increased dose comprises an incrementally increasing dose.

6 . The method of claim 1 , wherein the organ is selected from the group consisting of lungs, heart, liver, kidney, pancreas, intestine, thymus, and cornea.

7 . The method of claim 1 , wherein the initial dose, the increased dose, and the reduced dose are administered via an organ perfusion system or ventilation.

8 . A method to improve viability of an organ, the method comprising:

administering an initial dose of a nitric oxide gas to the organ;

administering an increased dose of the nitric oxide gas to the organ;

monitoring a methemoglobin level of the organ; and

administering a reduced dose of the nitric oxide gas when the methemoglobin level meets or exceeds a threshold methemoglobin level,

wherein the reduced dose of the nitric oxide gas is substantially equal to the initial dose of the nitric oxide gas,

wherein the initial dose, the increased dose, and the reduced dose do not exceed a concentration of 50 ppm of the nitric oxide gas, and

wherein the initial dose, the increased dose, and the reduced dose are administered for a combined time of twelve hours or less.

9 . The method of claim 8 , wherein the threshold methemoglobin level is about 5% to about 10%.

10 . The method of claim 8 , wherein the threshold methemoglobin level is about 10% to about 15%.

11 . The method of claim 8 , further comprising monitoring NO 2 and/or NO 3 levels.

12 . The method of claim 8 , wherein the increased dose comprises an incrementally increasing dose.

13 . The method of claim 8 , wherein the organ is selected from the group consisting of lungs, heart, liver, kidney, pancreas, intestine, thymus, and cornea.

14 . The method of claim 8 , wherein the initial dose, the increased dose, and the reduced dose are administered via an organ perfusion system or ventilation.

15 . A method to improve viability of an organ, the method comprising:

administering an initial dose of nitric oxide gas to the organ;

administering an increased dose of the nitric oxide gas to the organ;

monitoring a level of a nitric oxide marker of the organ; and

administering a reduced dose of the nitric oxide gas when the level of the nitric oxide marker meets or exceeds a threshold nitric oxide marker level,

wherein the initial dose, the increased dose, and the reduced dose do not exceed a concentration of 50 ppm of the nitric oxide gas, and

wherein the initial dose, the increased dose, and the reduced dose are administered for a combined time of twelve hours or less.

16 . The method of claim 15 , wherein the nitric oxide marker includes methemoglobin.

17 . The method of claim 16 , wherein the threshold nitric oxide marker level is about 5% to about 10%.

18 . The method of claim 16 , wherein the threshold nitric oxide marker level is about 10% to about 15%.

19 . The method of claim 15 , wherein the increased dose comprises an incrementally increasing dose.

20 . The method of claim 15 , wherein the nitric oxide marker includes NO 2 and/or NO 3 .
