Abstract

Provided are substrates and methods for detecting enzymatic activity of various lysosomal storage enzymes. The molecules may be used as substrates with improved solubility.

Claims (28)

1 . A substrate for detecting enzymatic activity of a lysosomal enzyme,

the substrate having the formula:

where A is D-glucose linked to O by a glycosidic bond;

where B1 is selected from the group consisting of: a methylene or an unsubstituted C 6 aryl;

B2 is a C 2 -C 7 chain comprising an amido, a urethane, an ester, a uriedo, a carbmamato, or a carbonyl bond to said molecule, wherein said C 2 -C 7 is unsubstituted, or wherein when said C 2 -C 7 comprises an alkyl or alkenyl chain, said B2 is unsubstituted or comprises a heteroatom substituent of N, O, or S, and wherein when said B2 is bonded by an amido said B2 is a C 2 -C 4 amido; and

B3 is selected from the group consisting of: an unsubstituted C 1 -C 20 alkyl, a heteroatom containing C 1 -C 20 alkyl; a C 1 -C 20 alkyl substituted with N, O, or S; an unsubstituted C 2 -C 20 alkenyl; a heteroatom containing C 2 -C 20 alkenyl; an C 2 -C 20 alkenyl substituted with N, O, or S; an unsubstituted C 2 -C 20 alkynyl; a heteroatom containing C 2 -C 20 alkynyl; a C 2 -C 20 alkynyl having substitution of N, O, or S; an C 6 -C 20 aryl; and an unsubstituted C 6 -C 20 heterocyclic containing a heteroatom of N, O or S;

or salts thereof.

2 . The substrate of claim 1 , wherein B1 is a methylene.

3 . The substrate of claim 1 , wherein B2 is a C 3 amido.

4 . The substrate of claim 1 , wherein B3 is a C 2 -C 20 alkenyl with a substituent of N, O, or S.

5 . The substrate of claim 1 , wherein: B1 is a methylene; B2 is a C 3 amido, and B3 is a C 2 -C 20 alkenyl with a substituent of O.

6 . A substrate composition for analysis of a plurality of enzymes associated with a lysosomal storage disorder, the substrate composition comprising a first substrate and

a second substrate different from said first substrate, said first substrate and said second substrate each independently and having the formula:

where A is a monosaccharide or a disaccharide linked to O by a glycosidic bond

B2 is a C 2 -C 7 chain comprising an amido, a urethane, an ester, a uriedo, a carbmamato, or a carbonyl bond to said molecule, wherein said C 2 -C 7 is unsubstituted, or wherein when said C 2 -C 7 comprises an alkyl or alkenyl chain, said B2 is unsubstituted or comprises a heteroatom or a substituent of N, O, or S, and wherein when said B2 is bonded by an amido, that said B2 is a C 2 -C 4 or C 6 amido; and

B3 is selected from the group consisting of: an unsubstituted C 1 -C 20 alkyl, a heteroatom containing C 1 -C 20 alkyl; a C 1 -C 20 alkyl substituted with N, O, or S; an unsubstituted C 2 -C 20 alkenyl; a heteroatom containing C 2 -C 20 alkenyl; an C 2 -C 20 alkenyl substituted with N, O, or S; an unsubstituted C 2 -C 20 alkynyl; a heteroatom containing C 2 -C 20 alkynyl; a C 2 -C 20 alkynyl having substitution of N, O, or S; an C 6 -C 20 aryl; and an unsubstituted C 6 -C 20 heterocyclic containing a heteroatom of N, O or S;

or salts thereof.

7 . The composition of claim 6 , wherein A in said first substrate is a D-glucose, and A in said second substrate is a D-galactose.

8 . The composition of claim 7 , wherein said first substate and said second substrate each have B2 of an unsubstituted C 2 -C 7 alkenyl comprising an amido, wherein B2 of said first substrate is a different B2 than said second substrate.

9 . The composition of claim 6 , wherein said first substrate is the substrate of claim 1 .

10 . The composition of claim 6 , wherein A is an aldohexose or a ketohexose.

11 . The composition of claim 6 , wherein A is a D-glucose or a D-galactose.

12 . The composition of claim 6 , wherein B1 a methylene in said first substrate and said second substrate.

13 . The composition of claim 6 , wherein B3 in said first substrate and said second substrate is a substituted C 2 -C 20 alkenyl with a substituent of N, O, or S.

14 . The composition of claim 6 , wherein A is a D-glucose or a D-galactose; B1 is a methylene; and B2 is a unsubstituted C 2 -C 7 comprising an amido.

15 . The substrate of claim 14 , wherein B3 is a substituted C 2 -C 20 alkenyl with a substituent of OH.

16 . The composition of claim 6 , wherein said first substrate has the formula:

17 . The composition of claim 16 , wherein said second substrate has the formula:
