Abstract

Systems and methods for filtering materials from biologic fluids are discussed. Embodiments may be used to filter cerebrospinal fluid (CSF) from a human or animal subject. The method may include the steps of withdrawing fluid comprising CSF, filtering the volume into permeate and retentate by passing the fluid through a tangential flow filter, and returning the permeate to the subject. During operation of the system, various parameters may be modified, such as flow rate.

Claims (52)

1 . A cerebrospinal fluid (CSF) filtration system, the system comprising:

a multi-lumen catheter configured for insertion into a CSF-containing space of a subject, the catheter having a withdrawal port and a return port;

a first tangential flow filter having a first inlet, a first permeate outlet, and a first retentate outlet, the first filter configured to separate withdrawn CSF into a first permeate and a first retentate;

a second filter having a second inlet coupled to the first retentate outlet, a second permeate outlet, and a second retentate outlet, the second filter configured to process the first retentate;

a combiner having a first input coupled to the first permeate outlet, a second input coupled to the second permeate outlet, and a combined output, the combiner configured to combine the first permeate and output from the second filter; and

a return flow path coupling the combined output to the return port of the multi-lumen catheter.

2 . The system of claim 1 , wherein the second filter comprises a dead-end filter or a depth filter configured to filter the first retentate into filtered retentate for combination with the first permeate.

3 . The system of claim 1 , wherein the second filter comprises a second tangential flow filter configured to separate the first retentate into a second permeate and a second retentate.

4 . The system of claim 3 , further comprising a waste collection system coupled to the second retentate outlet for collecting the second retentate.

5 . The system of claim 1 , wherein the combiner includes a check valve system configured to prevent backflow into the second filter.

6 . The system of claim 1 , further comprising:

a third filter having a third inlet coupled to the second retentate outlet when the second filter comprises a tangential flow filter; and

wherein the combiner is configured to combine the first permeate, the second permeate, and an output from the third filter.

7 . The system of claim 1 , wherein the second filter comprises a plurality of filters arranged in parallel, each filter having an inlet coupled to the first retentate outlet.

The system of claim 7 , wherein the plurality of filters comprises dead-end filters or depth filters that are self-regulating such that as one or more filter of the plurality of filters becomes clogged, increased pressure directs more of the first retentate to other filters in the plurality of filters.

9 . The system of claim 1 , further comprising:

a first flow control system positioned between the first retentate outlet and the second inlet, the first flow control system configured to regulate flow characteristics of the first retentate; and

a second flow control system positioned between the second retentate outlet and a waste collection point.

10 . The system of claim 1 , further comprising:

a first flow sensor positioned to measure flow rate of the first permeate;

a second flow sensor positioned to measure flow rate of output from the second filter; and

a controller configured to monitor the first flow sensor and the second flow sensor and adjust system parameters based on measured flow rates.

11 . A method for processing cerebrospinal fluid (CSF) using a dual-filter system, the method comprising:

withdrawing CSF from a CSF-containing space through a multi-lumen catheter;

filtering the withdrawn CSF through a first tangential flow filter to produce a first permeate and a first retentate;

processing the first retentate through a second filter to produce processed retentate;

combining the first permeate with the processed retentate in a combiner to form a combined fluid; and

returning the combined fluid to the CSF-containing space through the multi-lumen catheter.

12 . The method of claim 11 , wherein processing the first retentate comprises filtering the first retentate through a dead-end filter or depth filter.

13 . The method of claim 11 , wherein processing the first retentate comprises:

filtering the first retentate through a second tangential flow filter to produce a second permeate and a second retentate; and

wherein combining comprises combining the first permeate with the second permeate.

14 . The method of claim 13 , further comprising collecting the second retentate as waste material.

15 . The method of claim 11 , further comprising:

monitoring flow rates of the first permeate and the processed retentate;

calculating a total recovery rate based on the monitored flow rates; and

adjusting filtration parameters when the total recovery rate falls below a predetermined threshold.

16 . The method of claim 11 , wherein combining the first permeate with the processed retentate comprises preventing backflow into the second filter using a check valve in the combiner.

17 . The method of claim 11 , further comprising:

calculating a waste rate of the dual-filter system based on a difference between withdrawn CSF volume and returned combined fluid volume; and

modifying system parameters to maintain the waste rate below a predetermined threshold.

18 . A CSF processing system, the system comprising:

a primary filtration stage including a first tangential flow filter configured to separate CSF into a primary permeate and a primary retentate;

a secondary filtration stage including at least one secondary filter configured to process the primary retentate;

a combiner system configured to merge output from the primary filtration stage with output from the secondary filtration stage;

a flow regulation system configured to maintain predetermined flow ratios between the primary permeate and secondary filter output; and

a return system configured to deliver merged output to a subject.

The system of claim 18 , wherein the secondary filtration stage comprises multiple secondary filters operating in parallel, and wherein the flow regulation system comprises a flow regulator configured to distribute the primary retentate among the multiple secondary filters based on filter capacity status.

20 . The system of claim 18 , further comprising a waste rate monitoring system configured to:

measure volumes processed through the primary and secondary filtration stages;

calculate a system waste rate; and

provide feedback to the flow regulation system for maintaining the system waste rate below a waste threshold.
