Abstract

Aspects of the disclosure include methods for treating a myeloproliferative neoplasm. Methods according to certain embodiments include co-administering to a subject a Janus kinase (JAK) inhibitor and a telomerase inhibitor comprising an oligonucleotide and a lipid moiety linked to the 5′ and/or 3′ end of the oligonucleotide. Methods for inducing apoptosis of a myeloproliferative neoplasm cell by contacting the cell with an amount of a JAK inhibitor and a telomerase inhibitor sufficient to induce apoptosis are also described. Compositions having a JAK inhibitor and a telomerase inhibitor for practicing the subject methods are also provided.

128 . A kit comprising:

a first dosage form comprising a JAK inhibitor; and

a second dosage form comprising a telomerase inhibitor.

129 . The kit according to claim 128 , wherein the JAK inhibitor is ruxolitinib or a pharmaceutically acceptable salt thereof.

130 . The kit according to claim 128 , wherein the telomerase inhibitor is imetelstat or a pharmaceutically acceptable salt thereof.

131 . The kit according to claim 130 , wherein the telomerase inhibitor is imetelstat sodium.

132 . The kit according to claim 128 , wherein the first dosage form and the second dosage form are administered sequentially.

133 . The kit according to claim 132 , wherein the second dosage form is administered after the first dosage form.

134 . The kit according to claim 128 , wherein the first dosage form comprises about 5 mg/day to about 600 mg/day of the JAK inhibitor.

135 . The kit according to claim 128 , wherein the first dosage form comprises about 10 mg/day to about 400 mg/day of the JAK inhibitor.

136 . The kit according to claim 128 , wherein the first dosage form comprises about 10 mg/day to about 100 mg/day of the JAK inhibitor.

137 . The kit according to claim 128 , wherein the first dosage form comprises about 5 mg to about 40 mg the JAK inhibitor.

138 . The kit according to claim 128 , wherein the first dosage form comprises about 5 mg to about 25 mg the JAK inhibitor.

139 . The kit according to claim 128 , wherein the first dosage form comprises about 5 mg to about 20 mg the JAK inhibitor.

140 . The kit according to claim 128 , wherein the first dosage form comprises about 5 mg to about 15 mg the JAK inhibitor.

141 . The kit according to claim 128 , wherein the first dosage form comprises about 5 mg to about 10 mg the JAK inhibitor.

142 . The kit according to claim 128 , wherein the second dosage form comprises about 4 mg/kg to 10 mg/kg of the telomerase inhibitor.

143 . The kit according to claim 130 , wherein the second dosage form comprises about 8.9 mg/kg of imetelstat.

144 . The kit according to claim 131 , wherein the second dosage form comprises about 9.4 mg/kg of imetelstat sodium.

145 . The kit according to claim 128 , wherein the first dosage form is administered twice per day.

146 . The kit according to claim 128 , wherein the second dosage form is administered by intravenous infusion.

147 . The kit according to claim 128 , wherein the second dosage form is administered once every 4 weeks.
