Abstract

The present disclosure provides antibodies that specifically bind complement pathway component C1s. The present disclosure provides nucleic acids comprising nucleotide sequences encoding the anti-C1s antibodies; and host cells comprising the nucleic acids. The present disclosure provides compositions comprising the anti-C1s antibodies. The present disclosure provides methods of use of the anti-C1s antibodies.

21 . An antibody that specifically binds activated C1s and comprises:

a light chain comprising a light chain variable (VL) region and a light chain constant region,

wherein the VL region comprises SEQ ID NO: 22; and

a heavy chain comprising a heavy chain variable (VH) region and a heavy chain IgG4 constant region,

wherein the VH region comprises SEQ ID NO: 14,

wherein the heavy chain IgG4 constant region comprises proline, glutamic acid, leucine, and serine substitutions at amino acid residues corresponding to positions 108, 115, 308, and 314, respectively, relative to the IgG4 constant region sequence of SEQ ID NO: 52.

22 . The antibody of claim 21 , wherein the light chain constant region comprises SEQ ID NO: 45.

23 . The antibody of claim 21 , wherein the heavy chain IgG4 constant region comprises SEQ ID NO: 28.

24 . The antibody of claim 21 , wherein the light chain comprises SEQ ID NO: 30.

25 . The antibody of claim 21 , wherein the heavy chain comprises SEQ ID NO: 29.

26 . The antibody of claim 21 , wherein the light chain comprises SEQ ID NO: 30 and the heavy chain comprises SEQ ID NO: 29.

27 . The antibody of claim 21 , wherein the antibody comprises two light chains and two heavy chains, and wherein:

each light chain comprises: a VL region comprising SEQ ID NO: 22; and a light chain constant region; and

each heavy chain comprises: a VH region comprising SEQ ID NO: 14; and a heavy chain IgG4 constant region comprising proline, glutamic acid, leucine, and serine substitutions at amino acid residues corresponding to positions 108, 115, 308, and 314, respectively, relative to the IgG4 constant region sequence of SEQ ID NO: 52.

28 . The antibody of claim 27 , wherein the light chain constant region of each light chain comprises SEQ ID NO: 45.

29 . The antibody of claim 27 , wherein the heavy chain IgG4 constant region of each heavy chain comprises SEQ ID NO: 28.

30 . The antibody of claim 27 , wherein each light chain comprises SEQ ID NO: 30 and each heavy chain comprises SEQ ID NO: 29.

31 . The antibody of claim 21 , wherein the antibody is a bispecific antibody or a multispecific antibody.

32 . An immunoconjugate comprising the antibody of claim 21 .

33 . A pharmaceutical composition comprising: the antibody of claim 21 ; and a pharmaceutically acceptable excipient.

34 . A nucleic acid or set of nucleic acids encoding the antibody of claim 21 .

35 . A vector or a set of vectors comprising the nucleic acid or set of nucleic acids of claim 34 .

36 . A host cell comprising the nucleic acid or set of nucleic acids of claim 34 .

37 . A method of producing an antibody that specifically binds activated C1s, the method comprising culturing the host cell of claim 36 , so that the antibody is produced.

38 . A method of inhibiting a complement pathway in a subject in need thereof, the method comprising administering to the subject a pharmaceutically effective amount of the antibody of claim 21 .

39 . A method of treating a complement-mediated disease or disorder in a subject in need thereof, the method comprising administering to the subject a pharmaceutically effective amount of the antibody of claim 21 .
