Abstract

The present disclosure relates to compositions and methods for treating autism spectrum disorder (ASD) by restoring an ASD patient's gut microbiota. These methods can be used with ASD patient with or without ongoing gastrointestinal symptoms. Provided here is a method for ASD treatment in a subject in need thereof comprising or consisting essentially of administering a therapeutic composition comprising a fecal microbe or a fecal microbiota preparation to the subject. Also provided here is a method comprises administering an antibiotic to a human subject; subjecting the human subject to a bowel cleanse; and administering purified fecal microbiota to the human subject. Further provided are evaluation and quantitative characterization of patient symptom improvements upon treatment described here.

21 . A pharmaceutical composition comprising (a) a community of fecal bacteria from a stool, or portion thereof, of a human donor; and (b) a bacterial isolate comprising bacteria of the genus Lactobacillus , wherein the community of fecal bacteria does not comprise the bacterial isolate.

22 . The pharmaceutical composition of claim 21 , wherein the bacterial isolate is derived from a human different from the human donor.

23 . The pharmaceutical composition of claim 21 , wherein the bacterial isolate comprises cultured bacteria.

24 . The pharmaceutical composition of claim 21 , wherein the bacteria of the genus Lactobacillus comprise Lactobacillus leichmannii.

25 . The pharmaceutical composition of claim 21 , wherein the community of fecal bacteria is lyophilized.

26 . The pharmaceutical composition of claim 21 , wherein the bacterial isolate is lyophilized.

27 . The pharmaceutical composition of claim 21 , wherein the community of fecal bacteria has been derived without selection from the stool, or portion thereof, of the human donor.

28 . The pharmaceutical composition of claim 21 , for use in the treatment of autism spectrum disorder (ASD) in a subject in need thereof.

29 . The pharmaceutical composition of claim 28 , wherein the subject in need thereof has a gastrointestinal (GI) symptom.

30 . The pharmaceutical composition of claim 28 , wherein the subject in need thereof has no GI symptom.

31 . A method for treating an autism spectrum disorder (ASD) or a gastrointestinal symptom associated with ASD in a subject in need thereof, the method comprising: orally administering to the subject an induction dose of a pharmaceutical composition according to a first dosing schedule, wherein the pharmaceutical composition comprises (a) a community of fecal bacteria from a stool, or portion thereof, of a human donor, and (b) a bacterial isolate comprising bacteria of the genus Lactobacillus , wherein the community of fecal bacteria does not comprise the bacterial isolate, subsequently orally administering to the subject a maintenance dose of the pharmaceutical composition according to a second dosing schedule, wherein the second dosing schedule comprises administering the pharmaceutical composition to the subject for at least 8 weeks.

32 . The method of claim 31 , wherein the bacterial isolate is derived from a human different from the human donor.

33 . The method of claim 31 , wherein the bacterial isolate comprises cultured bacteria.

34 . The method of claim 31 , wherein the bacteria of the genus Lactobacillus comprise Lactobacillus leichmannii.

35 . The method of claim 31 , wherein the community of fecal bacteria is lyophilized.

36 . The method of claim 31 , wherein the bacterial isolate is lyophilized.

37 . The method of claim 31 , wherein the community of fecal bacteria has been derived without selection from the stool, or portion thereof, of the human donor.

38 . The method of claim 31 , wherein the second dosing schedule has an equal or lower dosing frequency compared to the first dosing schedule.

39 . The method of claim 31 , wherein the treating reduces severity of the impairment in social interaction related to ASD or the gastrointestinal symptom associated with ASD in the subject.

40 . The method of claim 31 , wherein the maintenance does is administered for at least 10 consecutive wecks.
