Abstract

Compositions, systems, and methods for the display of analytes such as biomolecules are described. Display of analytes is achieved by coupling of the analytes to displaying molecules that are configured to associate with surfaces or interfaces. Arrays of analytes may be formed from the described systems for utilization in assays and other methods.

Claims (33)

31 . A composition, comprising:

a) a solid support comprising a plurality of sites, wherein each site is separated by an optically resolvable distance from any other site of the plurality of sites;

b) a plurality of particles attached to the plurality of sites, wherein each site of the plurality of sites is attached to only one particle of the plurality of particles;

c) a plurality of polypeptides attached to the plurality of particles, wherein each particle of the plurality of particles is attached to only one polypeptide of the plurality of polypeptides, and wherein the plurality of polypeptides comprises at least 100 unique primary amino acid sequences; and

d) a plurality of detectable affinity reagents bound to polypeptides of the plurality of polypeptides, wherein detectable affinity reagents of the plurality of detectable affinity reagents are bound to at least 2 different polypeptides of the plurality of polypeptides.

32 . The composition of claim 31 , wherein the plurality of polypeptides comprises at least 10 3 unique primary amino acid sequences.

33 . The composition of claim 32 , wherein the plurality of polypeptides comprises at least 25% of all polypeptide species of a proteome.

34 . The composition of claim 31 , wherein the plurality of polypeptides comprises at least 2 unique proteoforms.

35 . The composition of claim 31 , wherein the plurality of polypeptides has a dynamic range of at least 10 4 .

36 . The composition of claim 31 , wherein each particle of the plurality of particles comprises a nucleic acid origami and a plurality of first coupling moieties.

37 . The composition of claim 36 , wherein each site of the plurality of sites comprises a plurality of second coupling moieties, wherein each site is attached to a particle of the plurality of particles by coupling of at least one second coupling moiety of the plurality of second coupling moieties to at least one first coupling moiety of the plurality of first coupling moieties.

38 . The composition of claim 36 , wherein the nucleic acid origami comprises a first face and a second face, wherein the plurality of first coupling moieties is attached to the first face, and wherein a polypeptide of the plurality of polypeptides is attached to the second face.

39 . The composition of claim 38 , wherein the first face is substantially opposite from the second face.

40 . The composition of claim 38 , wherein the second face further comprises a separating group, wherein the polypeptide of the plurality of polypeptides is attached to the separating group.

41 . The composition of claim 40 , wherein the separating group comprises a polymer linker, a nucleic acid linker, or a nanoparticle linker.

42 . The composition of claim 31 , wherein each particle is covalently attached to only one polypeptide of the plurality of polypeptides.

43 . The composition of claim 31 , wherein the plurality of detectable affinity reagents comprises a plurality of multivalent affinity reagents.

44 . The composition of claim 31 , wherein the at least 2 different polypeptides bound by the detectable affinity reagents individually comprise a same epitope bound by the detectable affinity reagents.

45 . The composition of claim 31 , wherein the at least 2 different polypeptides bound by the detectable affinity reagents comprise differing epitopes of a plurality of different epitopes bound by the detectable affinity reagents.

46 . The composition of claim 31 , wherein each site is separated from any other site by a distance of at least 500 nanometers.

47 . The composition of claim 31 , wherein each detectable affinity reagent of the plurality of detectable affinity reagents recognizes the at least 2 unique primary amino acid sequences of the 100 or more primary amino acid sequences.

48 . The composition of claim 31 , wherein each site of the plurality of sites comprises a well.

49 . A composition, comprising:

a) a solid support comprising a plurality of sites, wherein each site is separated by an optically resolvable distance from any other site of the plurality of sites;

b) a plurality of particles attached to the plurality of sites, wherein each site of the plurality of sites is attached to only one particle of the plurality of particles;

c) a plurality of polypeptides attached to the plurality of particles, wherein each particle of the plurality of particles is attached to only one polypeptide of the plurality of polypeptides, and wherein the plurality of polypeptides comprises at least 100 unique primary amino acid sequences; and

d) a plurality of detectable affinity reagents contacted to the solid support, wherein a fraction of the detectable affinity reagents is bound to polypeptides of the plurality of polypeptides.

50 . A composition, comprising:

a) a solid support comprising a plurality of wells, wherein each well is separated by an optically resolvable distance from any other well of the plurality of wells;

b) a plurality of particles attached to the plurality of wells, wherein each well of the plurality of wells is attached to only one particle of the plurality of particles;

c) a plurality of polypeptides attached to the plurality of particles, wherein each particle of the plurality of particles is attached to only one polypeptide of the plurality of polypeptides; and

d) a plurality of detectable affinity reagents contacted to the solid support, wherein a fraction of the detectable affinity reagents is bound to polypeptides of the plurality of polypeptides.
