Abstract

Compounds of Formulae (I)-(VII) or their pharmaceutically acceptable salts can inhibit the G12C, G12D, G12V, and/or G13D mutants of Kirsten rat sarcoma (KRAS) protein and are expected to have utility as therapeutic agents, for example, for treating cancer. The disclosure also provides pharmaceutical compositions which comprise compounds of Formulae (I)-(VII) or pharmaceutically acceptable salts thereof. The disclosure also relates to methods for use of the compounds or their pharmaceutically acceptable salts in the therapy and prophylaxis of cancer and for preparing pharmaceuticals for this purpose.

A method of treating cancer comprising administering a therapeutically effective amount of a compound or a pharmaceutically acceptable salt thereof to a subject in need of such treatment, wherein the cancer is pancreatic cancer, colorectal cancer or lung cancer, and wherein the compound is selected from the group consisting of:

30 . The method of claim 29 , wherein the cancer is pancreatic cancer.

31 . The method of claim 29 , wherein the cancer is colorectal cancer.

32 . The method of claim 29 , wherein the cancer is lung cancer.

33 . The method of claim 32 , wherein the lung cancer is a non-small cell lung carcinoma (NSCLC).

34 . The method of claim 32 , wherein the lung cancer is a small cell lung carcinoma.

35 . The method of claim 29 , further comprising administering an additional active agent to the subject.

36 . The method of claim 29 , wherein the cancer is mediated by a KRAS-G12C mutation, a KRAS-G12D mutation, a KRAS-G12V mutation, and/or a KRAS-G13D mutation.

37 . The method of claim 29 , wherein the compound is:
