Abstract

The disclosure features macrocyclic compounds, and pharmaceutical compositions and protein complexes thereof, capable of inhibiting Ras proteins, and their uses in the treatment of cancers.

Claims (51)

1 . A compound having the structure of Formula Int-1:

or a salt thereof, wherein:

Q is hydrogen, —B(OH) 2 ,

 or halogen;

Z is hydrogen or optionally substituted C 1 -C 6 alkyl;

Y is halogen, —B(OH) 2 ,

 -CO 2 H, —CO 2 —(C 1 -C 6 alkyl), or —CN;

E is

 and

PNG is a protecting group.

2 . The compound of claim 1 , wherein E is
