Abstract

The present invention relates to at least one novel chimeric, humanized or CDR-grafted anti-IL-6 antibodies derived from the murine CLB-8 antibody, including isolated nucleic acids that encode at least one such anti-IL-6 antibody, vectors, host cells, transgenic animals or plants, and methods of making and using thereof, including therapeutic compositions, methods and devices.

Claims (10)

1. Isolated nucleic acid molecules encoding an antibody or antigen-binding fragment thereof that binds and inhibits the activity of human IL-6, wherein said antibody or antigen-binding fragment comprise:

a human antibody heavy chain constant region and a heavy chain variable region comprising CDR1, CDR2 and CDR3, which comprise the amino acid sequences SEQ ID NO: 1, SEQ ID NO:2 and SEQ ID NO:3, respectively; and a human antibody light chain constant region and a light chain variable region comprising CDR1, CDR2 and CDR3, which comprise the amino acid sequences SEQ ID NO:4, SEQ ID NO:5 and SEQ ID NO:6, respectively.

The isolated nucleic acid molecules of claim 1 , wherein said isolated nucleic acid molecules comprise: a nucleotide sequence that encodes a human antibody heavy chain constant region and a heavy chain variable region comprising SEQ ID NO:7; and a nucleotide sequence that encodes a human antibody light chain constant region and a light chain variable region comprising SEQ ID NO:8.

3. A method for producing an IL-6 antibody or antigen-binding fragment, therof, comprising translating the nucleic acid molecules according to claim 1 under conditions in vitro, in vivo or in situ, such that the IL-6 antibody or antibody fragment is expressed in detectable or recoverable amounts.

4. A composition, comprising the nucleic acid molecules according to claim 1 and a carrier or diluent.

A method for producing an IL-6 antibody or antigen-binding fragment comprising transforming a host cell capable of expressing said antibody or antigen-binding fragment therof in recoverable amounts with the nucleic acid molecules according to claim 1 and expressing said antibody or antigen-binding fragment therof from said host cell.

6. The method according to claim 5 wherein said host cell is a mammalian cell, a plant cell or a yeast cell.

7. A vector, comprising the nucleic acid molecule according to claim 1 .

8. The vector according to claim 7 , wherein said vector comprises at least one promoter selected from the group consisting of a late or early SV490 promoter, a CMV promoter, an HSV tk promoter, a pgk (phosphoglycerate kinase) promoter, a human immunoglobulin promoter and an F-1 alpha promoter.

9. The vector according to claim 7 , wherein said vector comprises at least one gene or portion thereof selected from the group consisting of methotrexate (MTX), green fluorescent prototein (GFP), dihydrofolate reductase (DHFR), neomycin (G418), and glutamine synthestase (GS).
