Abstract

Novel methods allowing for the simple optical detection and electrochemical detection of doubled-straded oligonucleotides are disclosed. The methods are rapid, selective and versatile. Advantageously, they do not require any chemical reaction on the probes or on the analytes since they are based on different electrostatic interactions between cationic poly(3-alkoxy-4-methylthiophene) derivatives and single-stranded or doubled-stranded (hybridized) oligonucleotides.

Claims (50)

1. A thiophene monomer comprising the formula:

wherein:

a) m is an integer ranging from 2 to 3;

b) R* is a quaternary ammonium;

c) Y is an oxygen atom or a methylene; and

d) R 1 is a methyl group or a hydrogen atom.

2. The thiophene monomer of claim 1 , wherein:

a) m=3;

b) R* is + NEt 3 ;

c) Y is an oxygen atom; and

d) R 1 is a methyl group.

3. The thiophene monomer of claim 1 , wherein:

a) m is=2;

b) R* is

c) Y is an oxygen atom; and

d) R 1 is a methyl group.

4. The thiophene monomer of claim 1 , wherein:

a) m is=2;

b) R* is

c) Y is an oxygen atom; and

d) R 1 is a methyl group.

5. The monomer of claim 1 , wherein:

a) m is=2;

b) R* is

c) Y is a methylene group; and

d) R 1 is a hydrogen atom.

6. The monomer of claim 2 , having the formula:

10. A process for the preparation of the monomer of claim 2 , comprising:

a) reacting a molecule of formula 1:

with a reagent of formula 2:

in the presence of a copper halide, to produce a first reaction mixture comprising a compound of formula 3:

b) isolating said compound of formula 3 from said first reaction mixture;

c) reacting said compound of formula 3 with ethylbromide, to produce a second reaction mixture comprising said monomer; and

d) recovering said monomer from said second reaction mixture.

11. The process of claim 10 , wherein said copper halide is CuI.

12. A process for the preparation of the monomer of claim 3 , comprising:

a) reacting a compound of formula 4:

 with 1-methylimidazole, to produce a reaction mixture; and

b) recovering said monomer from said reaction mixture.

13. A process for the preparation of the monomer of claim 4 , comprising:

a) reacting a compound of formula 4:

with 1,2-dimethylimidazole, resulting in the formation of a reaction mixture; and

b) recovering said monomer from said reaction mixture.

14. A process for the preparation of the monomer of claim 5 , comprising:

a) reacting a compound of formula 6:

 with 1-methylimidazole, resulting in the formation of a reaction mixture; and

b) recovering said monomer from said reaction mixture.
