Substituted bicyclic amines for the treatment of diabetes
View Patent ↗Described herein are substituted bicyclic amines. In particular, described herein are substituted bicyclic amines that are effective as antagonists of SSTR5 and useful for the treatment, control or prevention of disorders responsive to antagonism of SSTR5, such as type 2 diabetes, insulin resistance, lipid disorders, obesity, atherosclerosis, metabolic syndrome, depression, and anxiety.
1. A compound of structural formula I:

Ar is an aryl or heteroaryl, wherein the aryl or heteroaryl are optionally substituted with one or more substituents independently selected from the group consisting of:

X is O, NH or OCH 2 ;
R 1 is SO 2 H, CONHC 1-10 alkyl, —C 1-10 alkoxycarbonyl, aroyl, heteroaroyl, aryl, heteroaryl, cycloalkyl or cycloheteroalkyl wherein the CONHC 1-10 alkyl, —C 1-10 alkoxycarbonyl, aroyl, heteroaroyl, aryl, heteroaryl, cycloalkyl or cycloheteroalkyl are optionally substituted with one or more substituents independently selected from the group consisting of:

—OC 1-10 alkyl,

2. The compound or salt of claim 1 , wherein Ar is phenyl.
3. The compound or salt of claim 1 , wherein Ar is substituted with:
CO 2 H,
—OC 1-10 alkyl,
aryl substituted with one to three halogen,
heteroaryl,
aryl,
aryl substituted with one to three C 1-10 alkyl,
C 1-10 alkyl, and
C 3-6 cycloalkyl.
4. The compound or salt of claim 1 , wherein R 1 is aroyl or heteroaroyl.
5. The compound or salt of claim 1 , wherein R 1 is heteroaryl.
6. The compound or salt of claim 1 , wherein R 1 is substituted with:
—OC 1-10 alkyl,
halogen,
SO 2 C 1-10 alkyl,
—NH 2 ,
—CONH 2 ,
—CO 2 H,
—C 1-10 alkoxycarbonyl,
—CO 2 C 1-10 alkylaryl,
—CF 3 ,
—OCHF 2 ,
C 1-10 alkyl-OH, and
C 1-10 alkyl.

9. A composition comprising a compound of claim 1 in combination with a pharmaceutically acceptable carrier.
10. A method of treating diabetes comprising administering to a patient in need thereof a therapeutic amount of a compound of claim 1 .
