Abstract

Antibodies and compositions against Insulin-Like Growth Factor-1 Receptor (IGF-1R) and uses thereof are provided herein and antibodies and pharmaceutical compositions that can be used in methods of treating or reducing the severity of thyroid eye disease (TED) in subjects in need thereof, including reducing proptosis and Clinical Activity Score (CAS) in said subjects, by inhibiting the activity of IGF-1R with such antibodies and compositions.

Claims (12)

1. A method of treating thyroid eye disease by inhibiting insulin-like growth factor I receptor (IGF-IR) activity in a subject in need thereof, the method comprising administering a pharmaceutical composition comprising an antibody for treating the thyroid eye disease to the subject in need thereof, wherein the antibody comprises a light chain variable region having the amino acid sequence of SEQ ID NO: 13 and a heavy chain variable region having the amino acid sequence of SEQ ID NO: 14.

2. The method of claim 1 , wherein the antibody comprises a light chain having an amino acid sequence of SEQ ID NO: 93 and a heavy chain having an amino acid sequence of SEQ ID NO: 92.

3. The method of claim 1 , wherein the pharmaceutical composition is administered intravenously.

4. The method of claim 1 , wherein treating the thyroid eye disease reduces proptosis in an eye in the subject.

5. The method of claim 4 , wherein the antibody comprises a light chain having an amino acid sequence of SEQ ID NO: 93 and a heavy chain having an amino acid sequence of SEQ ID NO: 92.

6. The method of claim 1 , wherein treating the thyroid eye disease reduces Clinical Activity Score (CAS) in the subject.

7. The method of claim 6 , wherein the antibody comprises a light chain having an amino acid sequence of SEQ ID NO: 93 and a heavy chain having an amino acid sequence of SEQ ID NO: 92.

8. The method of claim 1 , wherein treating the thyroid eye disease reduces proptosis by at least 2 mm and reduces clinical activity score (CAS) of the subject.

9. The method of claim 8 , wherein the antibody comprises a light chain having an amino acid sequence of SEQ ID NO: 93 and a heavy chain having an amino acid sequence of SEQ ID NO: 92.

10. The method of claim 1 , wherein treating the thyroid eye disease reduces the severity of diplopia in the subject.

11. The method of claim 10 , wherein the antibody comprises a light chain having an amino acid sequence of SEQ ID NO: 93 and a heavy chain having an amino acid sequence of SEQ ID NO: 92.

12. The method of claim 1 , wherein the pharmaceutical composition is administered at a dose of about 10 mg/kg of the antibody.
