Cells producing antibody compositions with increased antibody dependent cytotoxic activity
View Patent ↗The present invention relates to a cell for the production of an antibody molecule such as an antibody useful for various diseases having high antibody-dependent cell-mediated cytotoxic activity, a fragment of the antibody and a fusion protein having the Fc region of the antibody or the like, a method for producing an antibody composition using the cell, the antibody composition and use thereof.
1. An isolated fucosyltransferase knock-out host cell wherein when a gene encoding an antibody molecule is introduced in to said host cell, said host cell produces an antibody composition comprising the antibody molecule,
said host cell being a mammalian cell,
said antibody molecule comprising a Fc region comprising complex N-glycoside-linked sugar chains bound to the Fc region,
said sugar chains comprising a reducing end which contains an N-acetylglucosamine, wherein the sugar chains do not contain fucose bound to the 6 position of N-acetylglucosamine in the reducing end of the sugar chains.
2. The isolated host cell of claim 1 , wherein said host cell is a CHO cell.
3. The isolated host cell of claim 1 , wherein said host cell is a NS0 cell.
4. The isolated host cell of claim 1 , wherein said host cell is a SP2/0 cell.
5. The isolated host cell of claim 1 , wherein said host cell is a YB2/0 cell.
6. The isolated host cell of claim 1 , wherein said antibody molecule is an IgG antibody.
7. An isolated fucosyltransferase knock-out host cell comprising a gene encoding an antibody molecule, wherein said host cell produces an antibody composition comprising the antibody molecule,
said host cell being a mammalian cell,
said antibody molecule comprising a Fc region comprising complex N-glycoside-linked sugar chains bound to the Fc region,
said sugar chains comprising a reducing end which contains an N-acetylglucosamine, wherein the sugar chains do not contain fucose bound to the 6 position of N-acetylglucosamine in the reducing end of the sugar chains.
8. The isolated host cell of claim 7 , wherein said host cell is a CHO cell.
9. The isolated host cell of claim 7 , wherein said host cell is a NS0 cell.
10. The isolated host cell of claim 7 , wherein said host cell is a SP2/0 cell.
11. The isolated host cell of claim 7 , wherein said host cell is a YB2/0 cell.
12. The isolated host cell of claim 7 , wherein said antibody molecule is an lgG antibody.
