Abstract

Compounds of the general formula (I) in which the substituents R, R 1 , R 2 , R 3 , R 4 , R 5 and R 6 are each as defined in claim 1 have renin-inhibiting properties and can be used as medicaments.

Claims (31)

1. Compound of the formula

in which

R 1 is optionally substituted aryl;

R 2 is C 1 -C 8 -alkyl, C 2 -C 8 -alkenyl or C 3 -C 8 -cycloalkyl or is phenyl- or naphthyl-C 1 -C 4 -alkyl, each of which is unsubstituted or mono-, di- or trisubstituted by C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy, hydroxyl, C 1 -C 4 -alkylamino, N,N-di-C 1 -C 4 -alkylamino, halogen and/or trifluoromethyl;

R 3 is hydrogen, C 1 -C 4 -alkyl or C 1 -C 8 -alkanoyl;

R 4 is hydrogen, C 1 -C 4 -alkyl or C 1 -C 8 -alkanoyl;

R 5 are each independently hydrogen, C 1 -C 8 -alkyl or, together with the carbon atom to which they are bonded, a C 3 -C 8 -cycloalkylidene radical;

R 6 is hydrogen or hydroxyl;

R are each independently 1-4 radicals selected from;

hydrogen, halogen, C 1 -C 8 -alkyl, 3- to 8-membered cycloalkyl, polyhalo-C 1 -C 4 -alkyl, polyhalo-C 1 -C 4 -alkoxy, C 1 -C 4 -alkoxy-C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy-C 1 -C 4 -alkoxy-C 1 -C 4 -alkyl, 3- to 8-membered cycloalkoxy-C 1 -C 4 -alkyl, hydroxyl, C 1 -C 8 -alkanoyloxy-C 1 -C 4 -alkyl, hydroxy-C 2 -C 8 -alkyl, C 1 -C 4 -alkylthio-C 1 -C 4 -alkyl, C 1 -C 8 -alkanesulphonyl-C 1 -C 4 -alkyl, thiazolylthio-C 1 -C 4 -alkyl, thiazolinylthio-C 1 -C 4 -alkyl, imidazolylthio-C 1 -C 4 -alkyl, optionally N-oxidized pyridylthio-C 1 -C 4 -alkyl, pyrimidinylthio-C 1 -C 4 -alkyl, optionally partially hydrogenated pyridyl- or N-oxidopyridyl-C 1 -C 4 -alkyl, C 1 -C 4 -alkanesulphonylamino-C 1 -C 4 -alkyl, trifluoro-C 1 -C 8 -alkanesulphonylamino-C 1 -C 4 -alkyl, pyrrolidino-C 1 -C 4 -alkyl, piperidino-C 1 -C 4 -alkyl, piperazino-C 1 -C 4 -alkyl, N′-C 1 -C 4 -alkylpiperazino-C 1 -C 4 -alkyl, N′-C 2 -C 8 -alkanoylpiperazino-C 1 -C 4 -alkyl, morpholino-C 1 -C 4 -alkyl, thiomorpholino-C 1 -C 4 -alkyl, S-oxothiomorpholino-C 1 -C 4 -alkyl, S,S-dioxothiomorpholino-C 1 -C 4 -alkyl, cyano-C 1 -C 4 -alkyl, carboxy-C 1 -C 4 -alkyl, C 1 -C 4 -alkoxycarbonyl-C 1 -C 4 -alkyl, carbamoyl-C 1 -C 8 -alkyl, N-mono- or N,N-di-C 1 -C 4 -alkylcarbamoyl-C 1 -C 4 -alkyl, optionally by C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy, hydroxyl, C 1 -C 4 -alkylamino, di-C 1 -C 4 -alkylamino, halogen and/or trifluoromethyl mono-, di- or trisubstituted phenyl, optionally by C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy, hydroxyl, C 1 -C 4 -alkylamino, di-C 1 -C 4 -alkylamino, halogen and/or trifluoromethyl mono-, di- or trisubstituted naphthyl, hydroxy-C 2 -C 8 -alkoxy, halo-C 2 -C 8 -(hydroxy)alkoxy, C 1 -C 8 -alkanesulphonyl-C 1 -C 4 -(hydroxy)alkoxy, amino-C 1 -C 4 -alkyl, C 1 -C 4 -alkylamino-C 1 -C 4 -alkyl, N,N-di-C 1 -C 4 -alkylamino-C 1 -C 4 -alkyl, N—C 1 -C 4 -alkanoylamino-C 1 -C 4 -alkyl, C 1 -C 8 -alkoxycarbonylamino-C 2 -C 8 -alkyl, amino-C 1 -C 4 -alkoxy, C 1 -C 4 -alkylamino-C 1 -C 4 -alkoxy, N,N-di-C 1 -C 4 -alkylamino-C 1 -C 4 -alkoxy, C 1 -C 8 -alkanoylamino-C 1 -C 4 -alkoxy, C 1 -C 8 -alkoxycarbonylamino-C 2 -C 8 -alkoxy, C 1 -C 8 -alkanoyl-C 2 -C 4 -alkoxy which bears the alkanoyl group in higher than the α-position, C 1 -C 8 -alkoxy, 3- to 8-membered cycloalkoxy, C 2 -C 8 -alkenyloxy, 3- to 8-membered cycloalkoxy-C 1 -C 4 -alkoxy, C 1 -C 4 -alkoxy-C 1 -C 4 -alkoxy, C 1 -C 4 -alkoxy-C 2 -C 4 -alkenyl, C 2 -C 8 -alkenyloxy-C 1 -C 4 -alkoxy, C 1 -C 4 -alkoxy-C 2 -C 4 -alkenyloxy, C 2 -C 8 -alkenyloxy-C 1 -C 4 -alkyl, C 1 -C 4 -alkylthio-C 1 -C 4 -alkoxy, C 1 -C 8 -alkanesulphonyl-C 1 -C 4 -alkoxy, C 1 -C 4 -alkylthio-C 1 -C 4 -(hydroxy)alkoxy, optionally by C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy, hydroxyl, C 1 -C 4 -alkylamino, di-C 1 -C 4 -alkylamino, halogen and/or trifluoromethyl mono-, di- or trisubstituted phenyl-C 1 -C 4 -alkoxy, optionally by C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy, hydroxyl, C 1 -C 4 -alkylamino, di-C 1 -C 4 -alkylamino, halogen and/or trifluoromethyl mono-, di- or trisubstituted naphthyl-C 1 -C 4 -alkoxy, each of which is unsubstituted or mono-, di- or trisubstituted by C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy, hydroxyl, C 1 -C 4 -alkylamino, di-C 1 -C 4 -alkylamino, halogen and/or trifluoromethyl, polyhalo-C 1 -C 4 -alkoxy, optionally partially hydrogenated pyridyl- or N-oxidopyridyl-C 1 -C 4 -alkoxy, thiazolyl-C 1 -C 4 -alkoxy, optionally N-oxidized morpholino-C 1 -C 4 -alkoxy, thiazolylthio-C 1 -C 4 -alkoxy, thiazolinylthio-C 1 -C 4 -alkoxy, imidazolylthio-C 1 -C 4 -alkoxy, optionally N-oxidized pyridylthio-C 1 -C 4 -alkoxy, pyrimidinylthio-C 1 -C 4 -alkoxy, C 1 -C 8 -alkanesulphonylamino-C 1 -C 4 -alkoxy, trifluoro-C 1 -C 8 -alkanesulphonyl-C 1 -C 4 -alkoxy, pyrrolidino-C 1 -C 4 -alkoxy, piperidino-C 1 -C 4 -alkoxy, cyano-C 1 -C 4 -alkoxy, carboxy-C 1 -C 4 -alkoxy, C 1 -C 4 -alkoxycarbonyl-C 1 -C 4 -alkoxy, carbamoyl-C 1 -C 4 -alkoxy, N—C 1 -C 8 -alkylcarbamoyl-C 1 -C 4 -alkoxy, N-mono- or N,N-di-C 1 -C 4 -alkylcarbamoyl-C 1 -C 4 -alkoxy, carboxy-C 1 -C 4 -alkyl, C 1 -C 4 -alkoxycarbonyl-C 1 -C 4 -alkyl, carbamoyl-C 1 -C 8 -alkyl, N-mono- or N,N-di-C 1 -C 4 -alkylcarbamoyl-C 1 -C 4 -alkyl, carboxy-C 1 -C 4 -alkoxy, C 1 -C 4 -alkoxycarbonyl-C 1 -C 4 -alkoxy, carbamoyl-C 1 -C 8 -alkoxy, C 1 -C and its pharmaceutically acceptable salt.

Compound according to claim 1 , in which R 1 is aryl which is optionally mono- or polysubstituted by C 1 -C 8 -alkyl, cyano, hydroxyl, amino, C 1 -C 6 -alkylamino, N,N-di-C 1 -C 6 -alkylamino, C 2 -C 6 -alkenyl, C 2 -C 6 -alkynyl, C 0 -C 6 -alkylcarbonylamino, C 1 -C 6 -alkoxycarbonylamino, halogen, trifluoromethyl, C 1 -C 8 -alkoxy, optionally N-mono- or N,N-di-C 1 -C 8 -alkylated carbamoyl, optionally esterified carboxyl, aryl or heterocyclyl.

3. Compound according to claim 1 , in which R 1 is an aryl substituted by C 1 -C 6 -alkylenedioxy.

4. Compound according to claim 1 of the formula

in which

R 1 , R 2 , R 3 , R 4 , R 5 and R 6 are each as defined for the compound of the formula (I) and R′ and R″ are each independently as defined for R for the compound of the formula (I).

5. Compound according to of claim 1 of the formula

in which R, R 1 , R 2 , R 3 , R 4 and R 5 are each as defined for the compound of the formula (I).

6. Compound according to claim 1 , in which

R are each independently 1-4 radicals selected from;

hydrogen, halogen, C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy-C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy-C 1 -C 4 -alkoxy-C 1 -C 4 -alkyl, C 1 -C 8 -alkoxy and C 1 -C 4 -alkoxy-C 1 -C 4 -alkoxy.

7. Compound according to claim 1 , in which

R 1 is optionally substituted aryl,

R 2 is C 1 -C 8 -alkyl;

R 3 is hydrogen;

R 4 is hydrogen;

R 5 are each independently hydrogen or C 1 -C 8 -alkyl;

R 6 is hydrogen;

R are each independently 1-4 radicals selected from;

hydrogen, halogen, C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy-C 1 -C 4 -alkyl, C 1 -C 4 -alkoxy-C 1 -C 4 -alkoxy-C 1 -C 4 -alkyl, C 1 -C 8 -alkoxy and C 1 -C 4 -alkoxy-C 1 -C 4 -alkoxy.

8. Pharmaceutical preparation comprising a compound of the general formula (I) according to claim 1 , and also customary excipients.

9. Pharmaceutical combination in the form of a preparation or of a kit composed of individual components, consisting a) of a compound of the general formula (I) according to claim 1 and b) at least one drug form whose active ingredient has cardiovascular action.
