Abstract

The present invention is directed to an antibody or derivative thereof of human origin for inhibiting platelet aggregation, characterized in that it is effective by substantially exclusive binding to the activated state of platelet integrin receptor GPIIb/IIIa.

Claims (13)

1. An Antibody of human origin, wherein said antibody inhibits platelet aggregation and has a greater binding affinity to the activated state of platelet integrin receptor GPIIb/IIIa than to the inactive conformation of the platelet integrin receptor GPIIb/IIIa, wherein the antibody comprises the heavy and light chain variable domains of the amino acid sequence of SEQ ID NO: 159.

An antibody derivative, wherein said derivative is a bivalent or multivalent antibody construct comprising the variable domains of the heavy and light chains of an antibody of human origin, wherein said antibody of human origin inhibits platelet aggregation and has a greater binding affinity to the activated state of platelet integrin receptor GPIIb/IIIa than to the inactive conformation of the platelet integrin receptor GPIIb/IIIa, wherein said antibody derivative specifically binds to the activated state of platelet integrin receptor GPIIb/IIIa and inhibits platelet aggregation, wherein said antibody derivative comprises:

(i) a heavy chain variable domain comprising CDR1 and CDR2 of the heavy chain variable domain encoded by nucleic acids comprising nucleotides 4 to 399 of SEQ ID NO: 3 and CDR3 having the sequence as set forth in SEQ ID NO: 163 and a light chain variable domain comprising CDR1 and CDR2 of the light chain variable domain encoded by nucleic acids comprising nucleotides 427 to 810 of SEQ ID NO: 3 and CDR3 having the sequence as set forth in SEQ ID NO: 170 or SEQ ID NO: 171; or

(ii) a light chain variable domain encoded by nucleic acids comprising nucleotides 427 to 810 of SEQ ID NO: 3, wherein the CDR3 encoded by SEQ ID NO: 3 is replaced by a CDR3 having the sequence as set forth in SEQ ID NO: 170 or SEQ ID NO: 171; or

(iii) a heavy chain variable domain encoded by nucleic acids comprising nucleotides 4 to 399 of SEQ ID NO:3, wherein the CDR3 encoded by SEQ ID NO:3 is replace by a CDR3 having the sequence as set forth in SEQ ID NO: 163.

An Antibody of human origin which specifically binds to the activated state of platelet integrin receptor GPIIb/IIIa and inhibits platelet aggregation, wherein said antibody is a single chain antibody fragment or a Fab fragment or comprising the variable domains of the heavy and light chains of a single chain antibody fragment or a Fab fragment, wherein said antibody comprises the heavy and light chain variable domains of the amino acid sequence of SEQ ID NO: 159.

An Antibody of human origin which specifically binds to the activated state of platelet integrin receptor GPIIb/IIIa and inhibits platelet aggregation, wherein said antibody is a single chain antibody fragment or a Fab fragment or comprising the variable domains of the heavy and light chains of a single chain antibody fragment or a Fab fragment, wherein said antibody comprises

(i) a heavy chain variable domain comprising CDR1 and CDR2 of the heavy chain variable domain encoded by nucleic acids comprising nucleotides 4 to 399 of SEQ ID NO: 3 and CDR3 having the sequence as set forth in SEQ ID NO: 163 and a light chain variable domain comprising CDR1 and CDR2 of the light chain variable domain encoded by nucleic acids comprising nucleotides 427 to 810 of SEQ ID NO: 3 and CDR3 having the sequence as set forth in SEQ ID NO: 170 or SEQ ID NO: 171; or

(ii) a light chain variable domain encoded by nucleic acids comprising nucleotides 427 to 810 of SEQ ID NO: 3, wherein the CDR3 encoded by SEQ ID NO: 3 is replaced by a CDR3 having the sequence as set forth in SEQ ID NO: 170 or SEQ ID NO: 171; or

(iii) a heavy chain variable domain encoded by nucleic acids comprising nucleotides 4 to 399 of SEQ ID NO:3, wherein the CDR3 encoded by SEQ ID NO:3 is replace by a CDR3 having the sequence as set forth in SEQ ID NO: 163.

An antibody of human origin which specifically binds to the activated state of platelet integrin receptor GPIIb/IIIa and inhibits platelet aggregation and the binding site of said antibody is formed by a variable light chain immunoglobulin domain and a variable heavy chain immunoglobulin domain, wherein the variable light chain domain is encoded by nucleic acids comprising nucleotides 427 to 810 of SEQ ID NO:3 and the heavy chain variable domain is encoded by nucleic acids comprising nucleotides 4 to 399 of SEQ ID NO:3, wherein the CDR3 of the variable heavy chain domain is replaced by SEQ ID NO:163.

An antibody of human origin which specifically binds to the activated state of platelet integrin receptor GPIIb/IIIa and inhibits platelet aggregation and the binding site of said antibody is formed by a variable light chain immunoglobulin domain and a variable heavy chain immunoglobulin domain, wherein the variable light chain domain is encoded by nucleic acids comprising nucleotides 427 to 810 of SEQ ID NO:3, wherein the CDR3 of the variable light chain is replaced by SEQ ID NO:170 and the heavy chain variable domain is encoded by nucleic acids comprising nucleotides 4 to 399 of SEQ ID NO:3, wherein the CDR3 of the variable heavy chain domain is replaced by SEQ ID NO:163.

An antibody of human origin which specifically binds to the activated state of platelet integrin receptor GPIIb/IIIa and inhibits platelet aggregation and the binding site of said antibody is formed by a variable light chain immunoglobulin domain and a variable heavy chain immunoglobulin domain, wherein the variable light chain domain is encoded by nucleic acids comprising nucleotides 427 to 810 of SEQ ID NO:3, wherein the CDR3 of the variable light chain is replaced by SEQ ID NO:171 and the heavy chain variable domain is encoded by nucleic acids comprising nucleotides 4 to 399 of SEQ ID NO:3, wherein the CDR3 of the variable heavy chain domain is replaced by SEQ ID NO:163.
